چکیده

Mucoviscidosis is the most common genetic autosomal recessive disease in Caucasian populations, with an incidence of 1/2200 – 2500 live birth and a carrier frequency of 1 in 25 – 30 persons. The disease is caused by mutations of the CFTR gene (cystic fibrosis transductance conductance regulator), which is located on chromosome 7 in locus q31.2. The CFTR gene is a large gene (250 kpb) with 27 exons, which produces a 6.5 kpb RNA messenger, the final product being a transmembrane protein with 1480 aminoacids which form an AMPc-regulated ATP-dependent chloride channel1, 2. The CFTR protein, a low conductance chloride channel, is located in the apical membrane of epithelial cells found in pancreatic, biliary and intestinal ducts, in the tracheobronchial tubes, uriniferous tubules, genital

Prenatal Genetic Diagnosis and Genetic Counselling in Couples at High Risk for Mucoviscidosis (cystic Fibrosis)

Correspondence to: Zoran L. Popa, e-mail: [email protected], Clinic of Obstetrics and Gynecology III, Victor Babes University of Medicine and Pharmacy Timisoara, Romania SUMMARY: Mucoviscidosis is the most common genetic autosomal recessive disease in Caucasian populations, a potentially lethal disease and therefore prenatal genetic diagnosis is essential for couples with increased risk of h...

Analysis of CFTR Gene Mutations in Children with Cystic Fibrosis, First Report from North-East of Iran

Objective(s):  More than 1500 registered mutations in cystic fibrosis transmembrane regulator (CFTR) gene are responsible for dysfunction of an ion channel protein and a wide spectrum of clinical manifestations in patients with cystic fibrosis (CF). This study was performed to investigate the frequency of a number of well-known CFTR mutations in North Eastern Iranian CF patients. Material and...

Genetic characterisation of circulating fetal cells allows non-invasive prenatal diagnosis of cystic fibrosis.

OBJECTIVES Cystic fibrosis (CF) is an autosomal recessive disease due to mutations in the cystic fibrosis transmembrane conductance regulator (CFTR) gene. The purpose of this study was to develop a molecular method to characterise both paternal and maternal CFTR alleles in DNA from circulating fetal cells (CFCs) isolated by ISET (isolation by size of epithelial tumour/trophoblastic cells). ME...

The Spectrum of Mutations in 100 Thalassemic Carriers Referred to Ghaem Hospital of Mashhad

Abstract Background Thalassemia is common in the Iranian population, and it must be considered in the differential diagnosis of the microcytic hypochromic anemia. The molecular analysis of β-thalassemia is necessary for prenatal molecular diagnosis. Α-thalassemia caused by loss of function of either one of the two duplicated α-globin genes or in less frequent non deletion mutations mostly loc...

The Value of ARMS/PCR and RFLP/PCR In Prenatal Diagnostic Accuracy Of -Thalassemia

Background: It is estimated that about 3,000 pregnancies in Iran are at risk for b-thalassemia each year. Objective: To evaluate the diagnostic accuracy of combination of ARMS/PCR and  RFLP/PCR techniques in prenatal diagnosis of b-thalassemia.Methods: Sixty-seven b-thalassemia carrier families were enrolled in this study. To analyze b-globin gene, amplification refractory mutation system (ARMS...
