Prolactin Causes the Dissociation of Prolactin from Membrane Receptor in Lactating Mouse Mammary Action of High Prolactin Concentration Plasma Cell:
نویسنده
چکیده
In order to characterize the dissociation of receptor-bound prolactin (PRL) by mammary cells, cells were prepared from lactating mice by collagenase digestion and loaded with PRL. The dissociation reaction was performed in the presence of PRL. In the concentration range 1 ng/ml-1 μg/ml examined, PRL at higher than 10 ng/ml accelerated the dissociation of PRL in a concentration-dependent manner. The action of PRL on dissociation was completed within a short period. At the end of the 1 h-incubation period, the dissociation rate constant (k_,) was about 2 times larger in the presence of 1 μg/ml of PRL than in its absence. The action of PRL occurred predominantly at the level of the plasma membrane receptor. Mammary cells with greater PRL-binding capacities had larger k_1 in response to PRL. The present data showed that the dissociation of PRL from the receptor was influenced by the concentration of PRL and by the PRL-binding capacity of the cell. The rate of PRL-receptor interaction is expressed by the equation of k_1(PRL-bound receptors). It is probable that at exceedingly high levels of PRL, the PRLreceptor interaction occurs more frequently in the presence of PRL-dependent dissociation than in its absence.

Effect of pheromones on the plasma level of prolactin during pregnancy and lactating periods in female rat
Introduction: Pheromones play a great role in the reproductive and social behavior of animals. The main sources of pheromones are urine and paracrine secretions. Through the neuroendocrine system, prolactin is a safe parameter to measure and compare the effects of pheromones on the sexual, maternal and also lactating behavior. Methods: Female rats were divided into 19 groups (n=8). To measur...
متن کاملPectic Acid Effects on Prolactin Secretion in GH3/B6 Rat Pituitary Cell Line
Background: Pectic acid extracted from plants increases the secretion of prolactin (PRL) when injected intravenously into ewes or fed to rats. Fragments of ewe hypophysis and lactating rabbit mammary gland incubated in vitro in the presence of pectic acid secreted more PRL and caseins compared to the controls. However, it is not known whether pectic acid directly stimulates PRL secretion in pi...
متن کاملMaternal zinc deficiency raises plasma prolactin levels in lactating rats.
There is an inverse relation between zinc (Zn) intake and plasma prolactin in men and nonpregnant women. Whether a relation exists in lactating women is unknown, despite the potential consequences of perturbations in prolactin regulation on lactation performance. We examined the effects of low Zn intake on prolactin concentration, the prolactin regulatory pathway in the pituitary gland, and lac...
متن کاملP-7: Testosterone and Gonadotrophin Hormones Plasma Concentration-Induced Changes in Sertoli and Leydig Cells Number in Hyperprolactinemic Mature Male Mice
Background: Hyperprolactinemia is a particular concern in the treatment of schizophrenic patients. The side effects associated with high prolactin levels can have a negative impact on GnRH pulsatile secretion and, consequently LH, FSH and testosterone pulsatility and follows by, on special testicular cells that have these hormon’s receptors such as sertoli and leydig cells. Quantification the d...
متن کاملA receptor site for prolactin in lactating mouse mammary tissues.
Prolactin iodinated by lactoperoxidase method showed immunologically, electrophoretically and biolo9gically similar properties to native prolactin and possessed enough specific radioactivity for receptor studies. 1251-prolactin was incubated with mouse mammary tissues at 8 days of lactation. Both binding and release of 1251-prolactin depended on incubation time and temperature and were maximal ...
