چکیده

In this study, the determination of the efficiency of a pilot-scale fermentation process using corn steep liquor as a nitrogen source was attempted in order to produce lactic acid via batch culturing of Lactobacillus sp. RKY2. Using pure glucose, fermentation efficiency characteristics, such as final lactic acid, cell growth, yield, and productivity were not substantially influenced by the scale-up of the laboratory-scale fermentation from 2.5to 30and 300-litre scale fermentations. In all experiments, the content of lactic acid produced increased in a linear fashion with increases in the initial glucose concentration. In the experiments using wood hydrolyzate, both lactic acid productivity and cell growth were decreased as a result of the scaling-up of the fermentation. This might be attributed to the toxic chemicals contained in the wood hydrolyzates. However, in all experiments, lactic acid yields remained higher than 90 % with regard to the amount of glucose consumed. Therefore, lactic acid was successfully produced by the pilot-scale bioreactor scheme adopted in this study.

Cell Entrapment of Lactobacillus casei subsp. casei ATCC 39392 for Lactic Acid Production

In this study, lactic acid production by repeated batch fermentation using cell entrapped methods was compared. Barium alginate beads, agar gel and polyurethane foam cubes were employed as carriers to immobilize Lactobacillus casei subsp. casei for the purpose of L (+)-lactic acid production. Increasing concentrations of lactic acid during fermentation were better tolerated by barium alginate e...

Response Surface Methodology for Optimization of the Erythromycin Production by Fed-Batch Fermentation Using an Inexpensive Biological Nitrogen Source

A novel source of biological nitrogen produced by multi-strain fermentation was developed for erythromycin production. The nitrogen composition of corn steep liquor, biological nitrogen and soybean flour was optimized by using Response surface methodology (RSM) firstly. A Box-Behnken design was used to estimate the optimum nitrogen composition for the production of erythromycin as follows: corn...

Production of mannitol by Lactobacillus intermedius NRRL B-3693 in fed-batch and continuous cell-recycle fermentations

Improved fermentation processes were developed for the production of mannitol by a heterofermentative lactic acid bacterium (Lactobacillus intermedius NRRL B-3693). A fed-batch fermentation protocol overcame limitations caused by high substrate concentrations. The process was developed using corn steep liquor and glucose as inexpensive industrial nutrient sources, supplemented with a small amou...
