Moreover, after weight loss, no differences among PNPLA3 genotypes were found in ALT levels or in the prevalence of steatosis (p = 0.1 and p = 0.2, respectively). Both the relationships between DBMI SDS and DALT and between DW/Hr and DALT were analysed, based on the different PNPLA3 genotypes, and the slope of the relative regression lines were compared (Fig. 1). Homozygous children for the PNPLA3 minor allele (148M) showed a stronger correlation between DALT and DW/Hr than those carrying the other genotypes, as we observed in the comparison of regression lines (p = 0.002) (Fig. 1A). The Association of PNPLA3 Variants with Liver Enzymes in Childhood Obesity Is Driven by the Interaction with Abdominal Fat

BACKGROUND AND AIMS A polymorphism in adiponutrin/patatin-like phospholipase-3 gene (PNPLA3), rs738409 C->G, encoding for the I148M variant, is the strongest genetic determinant of liver fat and ALT levels in adulthood and childhood obesity. Aims of this study were i) to analyse in a large group of obese children the role of the interaction of not-genetic factors such as BMI, waist circumferenc...

Healthy PNPLA3 risk allele carriers present with unexpected body fat composition. A study of one thousand subjects.

INTRODUCTION The common PNPLA3 (adiponutrin) variant p.I148M represents a major genetic driver of progression in non-alcoholic fatty liver disease (NAFLD). NAFLD is commonly associated with traits of the metabolic syndrome, therefore it is mostly suspected in obese individuals. Here, we investigate the association between the PNPLA3 variant and anthropometric traits in a cohort of healthy indiv...

Effect of Kelussia odoratissima Mozaff extract on PNPLA3 gene expression in non-alcoholic fatty liver and control rats

Background and Aims: Non-alcoholic fatty liver disease (NAFLD) is associated with such symptoms as steatosis, fibrosis, and liver cirrhosis. Kelussia has attracted assiduous attention due to its protective effects on the liver. The PNPLA3 gene is mainly expressed in the liver and plays a major role in the degradation rate of hepatic triglycerides. Therefore, the present study aimed to assess th...

Comparison of The Effect of Ketogenic Diet and Low Caloric Diet On Weight Loss in Iranian Obese and Overweight Children

Abstract Overweight and obesity has increased in prevalence over the last two decades in many developed and developing countries including Iran. The aim of this present study was to compare the weight reduction effects of ketogenic and low calorie diets on overweight and obese Iranian children. Seventy-six overweight or obese children aged 9-16 years recruited from outpatient Ghaem Hospital, Nu...

The Frequent Adiponutrin (PNPLA3) Variant p.Ile148Met Is Associated with Early Liver Injury: Analysis of a German Pediatric Cohort

Introduction. The common adiponutrin (PNPLA3) variant p.Ile148Met is associated with liver injury. Here, we investigate the association of this polymorphism with hepatic and metabolic traits in a pediatric cohort. Patients and Methods. The study cohort comprised 142 German children (age 5-9 years, 98 overweight, 19 children with NAFLD). Results. Overweight children presented with increased seru...
