Functional Group Imaging by Adhesion AFM Applied to Lipid Monolayers
نویسنده
چکیده
Recently developed adhesion atomic force microscopy was used as a technique to map the spatial arrangement of chemical functional groups at a surface with a lateral resolution of 20 nm. The ratio of the adhesion forces for different functional groups can be compared with values determined from the known surface energies. This concept was demonstrated by mapping the adhesive interaction of domains in a phase-separated lipid monolayer with the AF'M tip. The ratio of the adhesion forces for both phases corresponds with the theoretical number for the CHz and CH3 groups.

21 Atomic Force Microscopy: Interaction Forces Measured in Phospholipid Monolayers, Bilayers, and Cell Membranes
Atomic force microscopy (AFM) is a powerful technique which is commonly used to image surfaces at the nanoscale and single-molecule level, as well as to investigate physical properties of the sample surface using a technique known as force spectroscopy. In this chapter, we review our recent research where we used AFM to investigate physical properties of phospholipid monolayers, bilayers, and c...
متن کاملCharacterizing Pilus-Mediated Adhesion of Biofilm-Forming E. coli to Chemically Diverse Surfaces Using Atomic Force Microscopy
Biofilms are complex communities of microorganisms living together at an interface. Because biofilms are often associated with contamination and infection, it is critical to understand how bacterial cells adhere to surfaces in the early stages of biofilm formation. Even harmless commensal Escherichia coli naturally forms biofilms in the human digestive tract by adhering to epithelial cells, a t...
متن کاملOdd–Even Effects in the Friction of Self-Assembled Monolayers of Phenyl-Terminated Alkanethiols in Contacts of Different Adhesion Strengths
We have studied the frictional properties of self-assembled monolayers (SAMs) of phenyl-terminated alkanethiols, C6H5(CH2)nSH (n = 13–16) on template-stripped gold. The friction force was measured with atomic force microscopy (AFM), and the magnitude of the adhesion was controlled by immersing the sliding contact in ethanol (giving low adhesion) or dry N2 gas (giving enhanced adhesion relative ...
متن کاملFunctional microdomains of glycolipids with partially fluorinated membrane anchors: impact on cell adhesion.
Functional microdomains of glycolipids were designed by mixing neoglycolipids with partially fluorinated alkyl (F-alkyl) chains and matrix lipids with alkyl chains. Fluorescence images of the mixed lipid monolayers at the air-water interface demonstrated that it is possible to control both size and distribution of the microdomains by means of the strong demixing of alkyl and F-alkyl membrane an...
متن کاملCustom AFM for X-ray beamlines: in situ biological investigations under physiological conditions.
A fast atomic force microscope (AFM) has been developed that can be installed as a sample holder for grazing-incidence X-ray experiments at solid/gas or solid/liquid interfaces. It allows a wide range of possible investigations, including soft and biological samples under physiological conditions (hydrated specimens). The structural information obtained using the X-rays is combined with the dat...
