چکیده
The purposes of this study were t o examine the biologic effects of the engagement of the interleukin-7 receptor (IL-7R) with recombinant human interleukin-7 (rhlL-7) in immunophenotypically distinct T-lineage acute lymphoblastic leukemia (ALL) blasts and t o elucidate the biochemical nature of the IL-7R-linked transmembrane signal in rhlL-7responsive T-lineage ALL blast populations. In the absence of costimulants, rhlL-7 stimulated the in vitro proliferation and colony formation of freshly isolated leukemic blasts from six t o eight T-lineage ALL patients with a mean plating efficiency of 196 -+ 53 (background subtracted) colonies/105 blasts plated. Stimulation of T-lineage ALL blasts with rhlL-7 resulted in markedly enhanced tyrosine phosphorylation of six distinct phosphoproteins with molecular weights of 57, 72, 98,123,150, and 190 Kd, and induced a rapid increase in the production of inositol-1,4,5-trisphosphate ( lns-1,4,5-P3). which

متن کاملI-7: Maternal Signalling to the Placenta
Background: Though it is well established that maternal blood-borne signals influence highly the growth of the placenta, the mechanisms are not known. In vitro trophoblast culture models are limited by an inability to reconstruct the polarised bilayer of the human hemochorial placenta. We have used a first trimester villous tissue explant system to investigate how growth factors interact with p...
متن کاملRole of protein kinase C in signal attenuation following T cell receptor engagement.
T lymphocyte activation through stimulation of the T cell receptor complex and co-stimulatory receptors is associated with acute tyrosine phosphorylation of intracellular proteins, which in turn mediate downstream signaling events that regulate interleukin-2 expression and cell proliferation. The extent of protein tyrosine phosphorylation is rapidly attenuated after only 1-2 min of stimulation ...
متن کاملJAK3 protein tyrosine kinase mediates interleukin-7-induced activation of phosphatidylinositol-3' kinase.
The interleukin-7 (IL-7) receptor is expressed throughout T-cell differentiation and, although lacking a tyrosine kinase domain, mediates tyrosine phosphorylation in T cells. We have identified IL-7-induced activation of three cyoplasmic tyrosine kinases in T cells, Jak1, Jak3, and the src-like kinase p56lck. Many members of the cytokine receptor superfamily activate the Jak protein tyrosine ki...
