What Determines the Redox Potential of Ferredoxins
نویسندگان
چکیده
For many cytochromes P450 (P450), the electrons are transferred from the reductase to the P450 via a [2Fe-2S] cluster ferredoxin (e.g. putidaredoxin, terpredoxin or adrenodoxin). A similar role is played by several plant-type ferredoxins to transfer electrons from photosystem I to NADP. Other ferredoxins like E. coli ferredoxin are probably involved in the biogenesis of iron-sulfur clusters. To understand what modulates the redox potential of the metal center in the different proteins, we have developed a method to calculate redox potentials from first principles. In calculations for Anabaena PCC7119 ferredoxin, we found that certain changes in protein structure upon a change in redox state have a strong influence on the redox potential. Preliminary results indicate that the same mechanism may be important for determining the redox potentials of other ferredoxins.

The redox potentials of the two-iron plant and algal ferredoxins. An electrostatic model.
The two-iron-sulphur co-ordination centre in plant and algal ferredoxins is considered as a collection of charged ions whose net negative charge is twice that of the one-iron-sulphur protein rubredoxin. Calculation of the electrostatic free-energy changes for reduction of the two types of proteins indicates that the redox potential of the two-iron-sulphur proteins should be more negative than t...
متن کاملMidpoint redox potentials of plant and algal ferredoxins.
Midpoint potentials of plant-type ferredoxins from a range of sources were measured by redox titrations combined with electron-paramagnetic-resonance spectroscopy. For ferredoxins from higher plants, green algae and most red algae, the midpoint potentials (at pH 8.0) were between --390 and --425 mV. Values for the major ferredoxin fractions from blue-green algae were less negative (between --32...
متن کاملRedox and functional analysis of the Rieske ferredoxin component of the toluene 4-monooxygenase.
Toluene 4-monooxygenase catalyzes the NADH- and O2-dependent hydroxylation of toluene to form p-cresol. The four-protein complex consists of a diiron hydroxylase, an oxidoreductase, a catalytic effector protein, and a Rieske-type ferredoxin (T4moC). Phylogenetic analysis suggests that T4moC is part of a clade specialized for reaction with diiron hydroxylases, possibly reflected in the conservat...

Two ferredoxins have been purified from N2-fixing Mycobacterium flavum. Ferredoxin I had a minimum molecular weight (by amino acid analysis) of 11 230 and contained 7.4 mol Fe and 7.0 mol S2per mol. The ratio of the absorbance at 400 nm to that at 275 nm was 0.7. Ferredoxin I1 had a minimum molecular weight of 13478 and contained 3.4 mol Fe and 3.2 mol S2per mol. The ratio of the absorbance at ...

The relative reactivities toward reduction by free flavin semiquinones of cytochromes (c-type cytochromes, cytochrome b5, c'-type cytochromes) iron-sulfur proteins (high-redox-potential ferredoxins, rubredoxins, low-redoxpotential ferredoxins), and blue copper proteins (plastocyanin, azurins) are shown to correlate with calculations of the solvent exposure of the various prosthetic groups. In t...
