چکیده
The free concentration of bacteriophage T4-coded gene 32 (single-stranded DNA binding) protein in the cell is autoregulated a t the translational level during T4 infection of Escherichia coli. The control of the synthesis of this protein reflects the following progression of net (cooperative) binding affinities for the various potential nucleic acid binding targets present : single-stranded DNA > gene 32 mRNA > other T4 mRNAs3double-stranded DNA. I n this paper we show t h a t the free concentration of gene 32 protein is maintained a t 2 to 3 pM, and use the measured binding parameters for gene 32 protein, extrapolated to intracellular conditions, to provide a quantitative molecular interpretation of this system of control of gene expression. These results are then further utilized to define the specific autoregu-latory binding sequence (translational operator site) on the gene 32 mRNA a uniquely unstructured finite binding lattice terminated by elements of secondary structure not subject t o melting by gene 32 protein a t the autoregulated concentration, and to predict how this site must differ from those found on other T4 messenger RNAs. It is shown t h a t these predictions are fully consistent with available T4 DNA sequence data The control of free protein concentration a s a method of genome regulation is discussed in terms of other systems t o which these approaches may apply.

Regulation of gene 32 expression during bacteriophage T4 infection of Escherichia coli.
The gene 32 protein of the bacteriophage T4 plays an important role in genetic recombination, DNA repair, and DNA replication; the protein functions in these processes by virtue of a strong binding capacity for single-stranded DNA. During infections of Escherichia coli by bacteriophage carrying amber of temperature-sensitive mutations in gene 32, the altered gene 32 protein (that is, the amber ...
متن کاملThe purification and characterization of gene 59 protein from bacteriophage T4.
Gene 59 of bacteriophage T4 is involved in genetic recombination and recombination-dependent DNA replication. We purified the protein encoded by gene 59 after cloning the gene in an expression vector. The molecular weight and the sequence of seven N-terminal amino acids of the protein were consistent with the nucleotide sequence of the gene (Hahn, S., Kruse, U., and Ruger, W. (1986) Nucleic Aci...
متن کاملEvaluation of Cell Penetrating Peptide Delivery System on HPV16E7 Expression in Three Types of Cell Line
Background: The poor permeability of the plasma and nuclear membranes to DNA plasmids are two major barriers for the development of these therapeutic molecules. Therefore, success in gene therapy approaches depends on the development of efficient and safe non-viral delivery systems. Objectives: The aim of this study was to investigate the in vitro delivery of plasmid DNA encoding HPV16 E7 gene...
متن کاملCloning and evaluation of gene expression and purification of gene encoding recombinant protein containing binding subunit of coli surface antigens CS1 and CS2 from Enterotoxigenic Escherichia coli
Background & Objective: Enterotoxigenic Escherichia coli (ETEC) is a major causative agent of diarrhea. Enterotoxins and the colonization factors (CFs) are major virulence factors in ETEC infections. The bacterium binds to the intestinal epithelial cell surface through colonization factors and produces enterotoxins that cause excessive fluid and electrolyte secretion in the lumen of the intesti...
متن کاملمهار بیان ژن GFP به وسیله تداخل RNA (RNAi) در دودمان سلولی کارسینومای جنینی P19
Introduction: RNA interference (RNAi) is a phenomenon of gene silencing that uses double-stranded RNA (dsRNA), specifically inhibits gene expression by degrading mRNA efficiently. The mediators of degradation are 21- to 23-nt small interfering RNAs (siRNA). The use of siRNAs as inhibitors of gene expression has been shown to be an effective way of studying gene function in mammalian cells. Ai...
