The carcinogenicity of p-monomethylaminoazobenzene in various diets and the activity of this dye relative to p-dimethylaminoazobenzene.

چکیده
In studies on the carcinogenic activity of compounds related to podimethylaminoazobenzene it has been observed that p-monomethylaminoazobenzene is about as active as the dimethyl compound (10, 17). More recently Giese, Miller, and Baumann (4) showed that when these compounds were fed ad libitum at either of two levels for 2 89 to 3 89 months, the monomethyl compound always caused a higher incidence of hepatomas than the dimethyl compound. However, rats fed p-monomethylaminoazobenzene voluntarily consumed more diet and, therefore, more of the carcinogen. Other studies (13) demonstrated that p-dimethylaminoazobenzene was reversibly demethylated in vivo to p-monomethylaminoazobenzene, and that the latter compound appeared to be irreversibly demethylated to p-aminoazobenzene. Since both methylated compounds are present in the animal body when either dye is fed, it is difficult to determine which is the closer to the true carcinogen. The present studies were designed with this question in mind. The effect of various diets on the carcinogenicity of p-monomethylaminoazobenzene was determined, and a more rigid comparison was made of the activity of the monomethyl and dimethyl dyes. The diets selected were of two types. In the first group were diets high in riboflavin or in which hydrogenated coconut oil was substituted for corn oil; these rations are known to inhibit the induction of tumors

متن کاملTHE CARCINOGENICITY OF CERTAIN DERIVATIVES OF p-DIMETHYLAMINOAZOBENZENE IN THE RAT
1. Eighteen known or possible metabolites of the hepatic carcinogen 4- (or p-) dimethylaminoazobenzene were tested for carcinogenic activity in the rat. Of these compounds only 4-monomethylaminoazobenzene, a known metabolite, proved to be active. Eight compounds, which appear to be metabolites of the dye, were inactive; these included 4-aminoazobenzene, 4'-hydroxy-4-monomethylaminoazobenzene, 4...
متن کاملCertain effects of dietary fats on the production of liver tumors in rats fed p-dimethylaminoazobenzene.
Hydrogenated coconut oil and its component fatty acids appear to be the most striking examples of dietary lipid inhibitors reported for the carcinogen p-dimethylaminoazobenzene. When these substances were fed to rats as the sole sources of lipid in synthetic diets containing the azo dye very few liver tumors, if any, appeared by 6 months (3, 4). If," however, a fat such as corn oil was fed inst...

In contrast to all previous results with many derivatives of 4-dimethylaminoazobenzene, the ingestion by the rat of 3-methoxy-4-aminoazobenzene or its N-monomethyl or N,N-dimethyl derivatives was found to induce high incidences of squamouscell carcinomas of the ear duct. $-Methoxy-4-aminoazobenzene also induced low incidences of adenocarcinomas in the small intestine and mammary gland. In addit...
متن کاملThe inhibition of the carcinogenicity of p-dimethyl-aminoazobenzene by certain detergents and the effect of diet on the levels of azo dyes in rat tissues.
It is well established that the rate at which liver tumors occur in rats fed p-dimethylaminoazobenzene can be altered greatly by dietary means. This subject has been reviewed through 1944 (15) and extended in more recent work (3~ 16, 17). The dietary inhibitors and accelerators that have been reported include members of the vitamin B complex, certain lipids, some sulfur-containing amino acids, ...
