چکیده
Cell adhesion molecules are thought to play an important role in guiding cell migration and localization in the development of the embryo and in organogenesis. In the immune system, cell adhesion molecules enhance the efficiency of specific receptor-dependent lymphocyte-accessory cell and lymphocyte-target cell interactions ; they are also important in leukocyteendothelial cell interactions and lymphocyte recirculation. Recent studies with monoclonal antibodies (MAb) that perturb antigen-receptor-dependent T-lymphocyte functions have defined a number of cell surface molecules that are associated with lymphocyte function (lymphocyte functio~ associated or LFA antigens) (Table 1). The antigens LFA-1, CD2, LFA3, CD8, and CD4 appear to enhance antigen-specific functions by acting as cell adhesion molecules. Further studies have shown that the LFA-1, CD2, and LFA-3 molecules are also important in antigen-independent T-lymphocyte adherence and function and that the LFA-1 molecule is important in the adherence and function of essentially all leukocyte cell types. This review focuses on LFA-1, CD2, and LFA-3. The role of CD4 and CD8 is reviewed by Littman in this volume. We discuss (a) the contributions of LFA-1, CD2, and LFA-3 to antigen-dependent and antigen-

Localization of the binding site on intercellular adhesion molecule-3 (ICAM-3) for lymphocyte function-associated antigen 1 (LFA-1).
Intercellular adhesion molecule 3 (ICAM-3; CD50) is the predominant counter-receptor on resting T cells and monocytes for the leukocyte integrin, lymphocyte function-associated antigen 1 (LFA-1; CD11a/CD18), and may play an important role in the initial stages of the T cell-dependent immune response. Deletion of individual immunoglobulin superfamily (IgSF) domains of ICAM-3 and ICAM-3 IgSF doma...
متن کاملCharacterization of the function of intercellular adhesion molecule (ICAM)-3 and comparison with ICAM-1 and ICAM-2 in immune responses
We have characterized the immunobiology of the interaction of intercellular adhesion molecule 3 (ICAM-3; CD50) with its counter-receptor, leukocyte function-associated antigen 1 (LFA-1; CD11a/CD18). Purified ICAM-3 supported LFA-1-dependent adhesion in a temperature- and cation-dependent manner. Activation of cells bearing LFA-1 increased adhesiveness for ICAM-3 in parallel to adhesiveness for ...
متن کاملIntercellular adhesion molecule 3, a third adhesion counter-receptor for lymphocyte function-associated molecule 1 on resting lymphocytes
Recent studies suggest that some T and B lymphocyte cell lines bind to the integrin lymphocyte function-associated molecule 1 (LFA-1) chiefly through a pathway independent of its two known counter-receptors, intercellular adhesion molecules (ICAMs)-1 and -2. A monoclonal antibody (mAb) was raised that, in combination with blocking mAb to ICAM-1 and ICAM-2, can completely inhibit binding of thes...
متن کاملSignalling and adhesive properties of the integrin leucocyte function-associated antigen 1 (LFA-1).
Leucocyte function-associated antigen 1 (LFA-1; also known as CDlldCD18) belongs to the family of integrins which is one of the most important families of cell adhesion molecules and includes receptors that bind to the extracellular matrix as well as receptors involved in cell-cell interactions. Recent findings have demonstrated that integrins are both adhesion receptors and signalling molecule...
متن کاملICAM-3 interacts with LFA-1 and regulates the LFA-1/ICAM-1 cell adhesion pathway
The interaction of lymphocyte function-associated antigen-1 (LFA-1) with its ligands mediates multiple cell adhesion processes of capital importance during immune responses. We have obtained three anti-ICAM-3 mAbs which recognize two different epitopes (A and B) on the intercellular adhesion molecule-3 (ICAM-3) as demonstrated by sequential immunoprecipitation and cross-competitive mAb-binding ...
