Transient Gene Expression in Protoplasts of Phaseolus vulgaris Isolated from a Cell Suspension Culture.

چکیده
A cell suspension culture of Phaseolus vulgaris cv Negro jamapa was established. Protoplasts isolated from this culture are suitable for transient gene expression studies after DNA transfer by either electroporation or treatment with polyethylene glycol. The optimized conditions for both methods were established, taking into consideration expression and cell viability. We have investigated two features of posttranscriptional gene regulation. We demonstrate that inclusion of the RNA leader sequence, omega (Omega), derived from tobacco mosaic virus, and a dicot intron in the leader region together enhance gene expression in this legume species about 10-fold.

Protoplast isolation, transient transformation of leaf mesophyll protoplasts and improved Agrobacterium-mediated leaf disc infiltration of Phaseolus vulgaris: tools for rapid gene expression analysis
BACKGROUND Phaseolus vulgaris is one of the most extensively studied model legumes in the world. The P. vulgaris genome sequence is available; therefore, the need for an efficient and rapid transformation system is more imperative than ever. The functional characterization of P. vulgaris genes is impeded chiefly due to the non-amenable nature of Phaseolus sp. to stable genetic transformation. T...
متن کاملSHOOT REGENERATION FROM SAFFRON PROTOPLASTS IMMOBILIZED IN Ca-ALGINATE BEADS
Saffron (Crocus sativus L.) protoplasts were isolated from the cells of a suspension culture or calli with a solution of Cellulase, Pectinase and Hemicellulase and embedded in Ca-alginate beads. They were cultured with or without nurse cells in MS medium supplemented with 2,4-D and 6-benzylaminopurine at 25°C. After several changes of medium, cell-clusters appeared on the surface of the Ca-algi...
متن کاملBlue-light-dependent osmoregulation in protoplasts of Phaseolus vulgaris Pulvini.
Blue light was found to induce shrinkage of the protoplasts isolated from first-leaf lamina pulvini of 18-day-old Phaseolus vulgaris. The response was transient following pulse stimulation, while it was sustainable during continuous stimulation. No apparent difference was found between flexor and extensor protoplasts. Protoplasts of the petiolar segment located close to the pulvinus showed no d...
متن کاملGrapevine protoplasts as a transient expression system for comparison of stilbene synthase genes containing cGMP-responsive promoter elements.
A method for preparing elicitor-responsive protoplasts from grapevine cells kept in suspension culture was established. The protoplasts were employed in order to perform transient gene expression experiments produced by externally added plasmids. Using the gene coding for bacterial beta-glucuronidase as the reporter gene, the transient expression under the control of various promoters of stilbe...
متن کاملSHOOT REGENERATION FROM PROTOPLASTS OF PENNISETUM AMERICANUM L (MIL CEREAL)
A finely dispersed cell suspension was obtained from embryogenic calli of two genotypes of pearl millet (Pennisetum americanum L. Mil cereal). Protoplasts isolated from both cell suspensions divided and produced globular embryoids and calli. On an agar medium, the embryoids were induced to form albino shoots or albino plantlet- like structures
