چکیده

Sinomenine is an alkaloid contained in the root of Sinomenium acutum Rehd. et Wilson (Fam. Menishermaceae), growing wild in southern Japan, and has long been used as a home remedy for neuralgia and rheumatism. It has a chemical structure similar to that of morphine, as shown left (1). Ishiwari (2, 3), who first isolated this alkaloid and studied its pharmacological actions, found that sinomenine caused depression of blood pressure by dilating peripheral blood vessels in mammals and acceleration of thoracic lymph flow in dogs. He (4) assumed that the foregoing therapeutic effect of this alkaloid was probably due to the increased circulation in the locus by such peripheral vascular effect.•š Augmentation of lymph rich in protein caused by sinomenine (5) is also observed in the case of histamine(6), as well as with peptone (7, 8) and ana

Electron microscope studies on the release of histamine from rat peritoneal mast cells.

The occurrence of histamine in the granules of mammalian mast cells has been established by combined bioassay and histological studies (RILEY and WEST, 1953), by chemical analysis of the granule fraction (HAGEN, BARRNETT and LEE, 1959) and further by fluorescence histochemistry for histamine (JUHLIN and SHELLEY, 1966; YAMASAKI, 1967; TAKAYA, 1969). Certain substances such as compound 48/80 (PAT...

Plasma histamine and coagulation time of the blood in dogs after administration of different histamine releasers.

Compound 48/80, sinomenine, tween 20 and polyvinylpyrrolidone (PVP) were injected intravenously to dogs, in doses producing similar degree of profound hypotension, and changes in the plasma histamine content and coagulation time were followed on the blood from the femoral artery. After the injection of 48/80 or sinomenine plasma histamine rose rapidly and markedly, attaining its maximum within ...

Histamine release inhibition in anti-inflammatory mechanism.

Rats were depleted of skin histamine by more than 80 % by intraperitoneal injections of sinomenine with daily increasing doses for 6 days. In these rats, egg-white edema induced in the hind paws was inhibited by 68 % of control. The weight of the wall of granuloma pouch made by croton oil was also evidently smaller in the rat treated similarly with sinomenine than that of control. This suggests...

ENDOGENOUS RELEASE OF OPIATES BY REPETITIVE ELECTRICAL FIELD STIMULATION IN THE GUINEA-PIG AND RAT ILEAL LONGITUDINAL MUSCLE

The effect of repetitive electrical field stimulation and the response of the guinea-pig and rat ileal longitudinal muscle to single pulse stimulations was examined. Single pulse field stimulation produced twitch contraction which was inhibited by repetitive field stimulation (10 Hz, 40V, 0.5 msec for 5 m). This inhibition was largely, though never completely, reversed by naloxone. Contrac...

Controlled release of optimized electroporation enhances the transdermal efficiency of sinomenine hydrochloride for treating arthritis in vitro and in clinic

Sinomenine hydrochloride (SH) is an ideal drug for the treatment of rheumatoid arthritis and osteoarthritis. However, high plasma concentration of systemically administered SH can release histamine, which can cause rash and gastrointestinal side effects. Topical delivery can increase SH concentration in the synovial fluid without high plasma level, thus minimizing systemic side effects. However...
