Topoisomerase II isoforms are required for HIV-1 reverse transcription

چکیده
Background Human topoisomerase II is present in two forms 170kDa a and 180 kDa b isoform. Early reports have indicated that the topoII poisons and antisense RNA can block viral replication. The objective of present study is to evaluate specific requirement of topoII isomerases in promoting HIV replication and to characterize the HIV-1 replication intermediates to study molecular action of the topoII a or/and b isoforms.

Reverse Transcriptase and Cellular Factors: Regulators of HIV-1 Reverse Transcription
There is ample evidence that synthesis of HIV-1 proviral DNA from the viral RNA genome during reverse transcription requires host factors. However, only a few cellular proteins have been described in detail that affect reverse transcription and interact with reverse transcriptase (RT). HIV-1 integrase is an RT binding protein and a number of IN-binding proteins including INI1, components of the...

Objective: In this paper, we have introduced topoisomerase inhibitors, mechanism of action and types of them. DNA topoisomerases are ubiquitous enzymes that catalyze essential enzymes to solve the topological problems accompanying key nuclear processes such as DNA replication, transcription, repair and chromatin assembly by introducing temporary single or double strand breaks in the DNA. Result...

متن کاملC-Terminal regions of topoisomerase IIα and IIβ determine isoform-specific functioning of the enzymes in vivo
Topoisomerase II removes supercoils and catenanes generated during DNA metabolic processes such as transcription and replication. Vertebrate cells express two genetically distinct isoforms (alpha and beta) with similar structures and biochemical activities but different biological roles. Topoisomerase IIalpha is essential for cell proliferation, whereas topoisomerase IIbeta is required only for...
متن کاملIn Vitro Topoisomerase II Inhibitory and Apoptotic Activities of Novel 3,5 Disubstituted Thiophene-2-Carboxyl
Topoisomerases (topoII) are crucial enzymes involved during DNA replication, repair and transcription. Recent studies have shown topo II as an interesting target for cancer therapy. In the current study we have synthesized and characterized few thiophene derivatives and determined their antiproliferative activity and apoptosis studies in cancer cell lines. Biochemical analysis showed that thiop...
