Bile Salt Transport after Selective Damage to Acinar Zone 3 Hepatocytes by Bromobenzene in the Rat, GUMUCIO, J. J., KATZ, M. E., MILLER, D. L., BALABAUD, C. P., GREENFIELD, J. M., AND WAGNER, R. M. (1979). Toxicol. Appl. Pharmacol. 50, 77-85. To quantitate the contribution of acinar zone 3 hepatocytes (centrilobular cells) to the transport of physiological concentrations of taurocholate, selective acinar zone 3 cell damage was induced in female rats by the administration of bromobenzene. Forty-eight hours later, livers were perfused in situ. Oxygen consumption, gluconeogenesis, transport of [%]taurocholate into bile, bile flow and capacity for removal of indocyanine green were assessed. The removal of single doses of [i4C]taurocholate injected into the portal vein at loads of 0.025 and 0.11 pmol/kg was then measured. Bromobenzene-treated livers removed 8&85 % of the injected dose while control livers removed about 98 %. The percentage of the injected dose removed by each group at each load of taurocholate remained fairly constant. In separate experiments, [‘%]taurocholate was administered in successive single injections at loads between 0.20 and 75 pmol/kg. The percentage of the injected dose removed remained constant and was similar to that observed at lower loads of taurocholate until a dose of 1 .O pmol/kg was used. At higher loads, the percentage of the injected dose removed decreased progressively, suggesting that the uptake process was reaching saturation. These results showed that acinar zone 3 hepatocytes contributed to the removal of 13-l 8 % of a physiological load of taurocholate. Furthermore, a 50 % decrease in bile flow was observed after bromobenzene administration. This suggested that damage to .the centrilobular area is followed by alterations in bile flow and by the passage of at least 13-18x of the bile salts reaching the acinus into the general circulation. In addition, taurocholate removal after carbon tetrachloride-induced damage to the centrilobular area was studied. Twelve to eighteen percent of the injected dose of [‘%]taurocholate was found in the hepatic vein effluent, values similar to the results obtained after bromobenzeneinduced damage.

Bromobenzene-induced zonal necrosis in the hepatic acinus.
The time course and acinar distribution of bromobenzene-induced hepatic necrosis was studied in the rat. Cellular damage, lipid infiltration, and changes in glycogen deposits were investigated by light microscopy 6, 16, 24, and 48 hr after bromobenzene (BZ) administration. Concomitantly, ultrastructural changes were followed by electron microscopy in each zone of the acinus. To insure accurate ...
متن کاملExtrahepatic obstructive cholestasis reverses the bile salt secretory polarity of rat hepatocytes.
To elucidate the consequences of extrahepatic cholestasis on the structure and function of hepatocytes, we studied the effects of bile duct ligation on the turnover, surface distribution, and functional activity of the canalicular 100-kD bile salt transport protein (cBSTP). Basolateral (blLPM) and canalicular (cLPM) liver plasma membrane vesicles were purified to the same degree from normal and...
متن کاملSmall hepatocytes in culture develop polarized transporter expression and differentiation.
Rat small hepatocytes have been shown to proliferate in culture and to form organoids with differentiated hepatocytes in vitro. To evaluate the degree of polarized transporter differentiation of rat small hepatocytes during 9 weeks of culturing, we studied the time-dependent expression and subcellular localization of the major bile salt and organic anion transport systems of hepatocytes [i.e. t...
متن کاملRitonavir, saquinavir, and efavirenz, but not nevirapine, inhibit bile acid transport in human and rat hepatocytes.
Human immunodeficiency virus-infected patients on antiretroviral drug therapy frequently experience hepatotoxicity, the underlying mechanism of which is poorly understood. Hepatotoxicity from other compounds such as bosentan and troglitazone has been attributed, in part, to inhibition of hepatocyte bile acid excretion. This work tested the hypothesis that antiretroviral drugs modulate hepatic b...
متن کاملThe role of lipid peroxidation in the toxicity of foreign compounds to liver cells.
The peroxidation of polyunsaturated fatty acids present in the membrane lipids has been proposed as the mechanism by which a number of foreign compounds produce structural tissue injury [1-3]. For certain chemicals, such as bromotrichloroethane and carbon tetrachloride, it is clear that lipid peroxidation may have a critical role in bringing about the tissue damage [3-5]. However, in respect to...
