چکیده

Obesity is an epidemic in the US and an established risk factor for breast cancer incidence and morbidity. In obese patients breast cancer is commonly more aggressive and associated with poor prognosis. The goal of our research is to develop safe and efficient novel therapies to treat aggressive breast tumors, like those in obese patients. To enhance drug uptake we intend to develop electro-endocrine therapy, an electrically mediated hormone (endocrine) delivery system. In this study, we optimize parameters for electrical pulses and tamoxifen (anti-hormone drug) dosages in an in vitro model of breast cancer, as these parameters vary for each cell type. For this purpose, MCF-7 human breast adenocarcinoma cells were used. Results of our experiments show that treatment of electroporation combined with tamoxifen has an enhanced effect on suppressing tumor cell growth compared to tamoxifen treatment alone. The longterm goal is to translate application of this method to clinical practice. r 2007 Elsevier B.V. All rights reserved.

CDK4/6 inhibition in luminal breast cancer

Endocrine therapy represents the basis for the treatment of estrogen receptor-positive breast cancer, but several tumors harbor intrinsic resistance and acquired resistance to endocrine therapy is inevitable in metastatic disease. Combination strategies of endocrine therapy with targeted agents are aimed to overcome endocrine resistance. The selective CDK4/6 inhibitor palbociclib has shown prom...

Activation of PI3K/Akt/mTOR signaling in the tumor stroma drives endocrine therapy-dependent breast tumor regression

Improved efficacy of neoadjuvant endocrine-targeting therapies in luminal breast carcinomas could be achieved with optimal use of pathway targeting agents. In a mouse model of ductal breast carcinoma we identify a tumor regressive stromal reaction that is induced by neoadjuvant endocrine therapy. This reparative reaction is characterized by tumor neovascularization accompanied by infiltration o...

mTOR inhibitors in the treatment of breast cancer.

The phosphatidylinositol 3-kinase/mammalian target of rapamycin (PI3K/mTOR) pathway is commonly dysregulated in breast cancer. In preclinical studies, hyperactivation of the PI3K pathway has been linked to resistance to both endocrine therapy and trastuzumab (Herceptin). Rapalogs, agents that primarily inhibit mTOR-raptor complex 1, have been studied in combination with endocrine therapy to ove...

Adjuvant systemic therapy for breast cancer.

The first procedure in primary breast cancer is usually the surgical excision of the tumor. However, a medical therapy is necessary in almost all patients to treat the systemic component of the disease. Which medical approach is recommended depends on the biology of the tumor itself. Endocrine-responsive tumors must be treated by an endocrine therapy according to their menopausal status. In HER...

Protein Kinase C Alpha in Breast Cancer: A Focus on Endocrine Resistant and Triple Negative Breast Cancer

Breast cancer remains the most common cancer in women worldwide and the second most common cancer among women in the U.S. The optimal treatment for patients is based on the expression of the estrogen receptor α (ER), progesterone receptor (PR) and HER2 as assessed by immunohistochemical methods. Patients with tumors that express ER are treated with endocrine therapy including tamoxifen or aroma...
