چکیده

By E. KODICEK, M.D., Ph.D., AND K. J. CARPENTER, MA., Ph.D. I N OUR previous paper’ we described the development of a macrocytic normochromic anemia in rats fed on synthetic diets containing i per cent sulfasuxidine, and maintained in a state of chronic pteroylglutamic acid (PGA) deficiency. The blood picture of these rats was similar to the nutritional anemias of other species, caused by a PGA deficiency. Although sulfasuxidine was effective in producing anemia, it caused certain pathologic changes, such as the deposition of stones in the urinary tract, which were due to the slightly soluble sulfasuxidine being absorbed in small amounts into the blood stream and subsequently deposited during excretion. Other sulfonamides, namely the less soluble sulfaguanidine, phthalylsulfathiazole and the more soluble sulfathiazole, were therefore studied in a further series of experiments. Sulfaguanidine and phthalylsulfathiazole have been successfully used by a number of workers.’’ Sulfaguanidine, however, was found by Slater#{176} to be toxic for rats and had to be replaced by sulfasuxidine. The more soluble sulfathiazole, in

Studies on the Relationship of Pteroylglutamic Acid to the Growth of Psittacosis Virus (strain 6bc)

p-Aminobenzoic and pteroic acids antagonize the inhibition of the growth of psittacosis virus (strain 6BC) by sulfadiazine in a competitive manner. Pteroylglutamic acid exerts a non-competitive type sulfonamide antagonism. The effects of certain analogues of pteroylglutamic acid and of pteroic acid on the growth of psittacosis virus and its inhibition by sulfadiazine are reported. The implicati...

The Kinetics of Iron Metabolism in Swine with Various Experimentally Induced Anemias

Ferrokinetic studies were performed on 3 swine given phenylhydrazine, 3 swine deficient in pyridoxine, and 3 swine deficient in pteroylglutamic acid. Body surface radioactivity was measured in 2 pteroylglutamic acid-deficient animals. In the animals given phenylhydrazine, the mean erythrocyte survival time was 5 days. The plasma iron turnover rate was increased about fourfold, and the rate of e...

The Effect of Alpha-Lipoic Acid on Learning and Memory Deficit in a Rat Model of Temporal Lobe Epilepsy

Introduction: Epilepsy is a chronic neurological disorder in which patients experience spontaneous recurrent seizures and deficiency in learning and memory. Although the most commonly recommended therapy is drug treatment, some patients do not achieve adequate control of their seizures on existing drugs. New medications with novel mechanisms of action are needed to help those patients whose sei...

The requirement of the normal rat for pteroylglutamic acid.

Many workers on the subject of pteroylglutamic-acid (PGA) deficiency in the rat have obtained data about the requirement of the rat for this vitamin; but so far the emphasis has been on the therapeutic dose required to correct a given deficiency rather than on the amount necessary for maintenance. Figures for the normal requirement are needed as a guide in the preparation of certain purified di...

INTERRELATIONSHIP BETWEEN p-AMINOBENZOIC ACID AND PTEROYLGLUTAMIC ACID AS GROWTH FACTORS FOR LACTOBACILLI*

The elucidation of the structure of pteroylglutamic acid (PGA) (Lactobacillus casei factor, vitamin B,) has shown that p-aminobenzoic acid (PABA) is a component of the PGA molecule (1). Since earlier work has treated PABA and PGA as distinct growth factors for bacteria (2), it is now necessary to study the relationship in the utilization of these substances by bacteria. It has been shown that m...
