Myristic acid increases ∆6-desaturase activity in cultured rat hepatocytes

چکیده
In order to study the effects of saturated fatty acids on ∆6-desaturase activity, rat hepatocytes in primary culture were incubated with lauric (C12:0), myristic (C14:0) or palmitic (C16:0) acids. After optimization, the standard in vitro conditions for the measurement of ∆6desaturase activity were as follows: 60 μmol·L–1 α-linolenic acid (C18:3n-3), reaction time of 20 min and protein content of 0.4 mg. Data showed that cell treatment with 0.5 mmol·L–1 myristic acid during 43 h specifically increased ∆6-desaturase activity. This improvement, reproducible for three substrates of ∆6-desaturase, i.e. oleic acid (C18:1n-9), linoleic acid (C18:2n-6) and α-linoleic acid (C18:3n3) was dose-dependent in the range 0.1–0.5 mmol·L–1 myristic acid concentration. myristic acid / ∆6-desaturase / saturated fatty acids / cultured rat hepatocytes

Although it is rapidly metabolized in cultured rat hepatocytes, lauric acid is used for protein acylation.
This study was designed to examine the metabolic fate of exogenous lauric acid in cultured rat hepatocytes, in terms of both lipid metabolism and acylation of proteins. Radiolabeled [14C]-lauric acid at 0.1 mM in the culture medium was rapidly taken up by the cells (94.8 +/- 2.2% of the initial radioactivity was cleared from the medium after a 4 h incubation) but its incorporation into cellular...
متن کاملFatty acid metabolism in hepatocytes cultured with hypolipidaemic drugs. Role of carnitine.
The direct effects of clofibrate analogues on carnitine acyltransferase activities and fatty acid metabolism were studied in cultured hepatocytes. Rat hepatocytes cultured with bezafibrate or ciprofibrate (0.1-10 micrograms/ml) for 48 h had increased activities of carnitine acetyltransferase (CAT; 4-6-fold) and carnitine palmitoyltransferase (CPT; 12-34%). The increase in CAT was higher in hepa...
متن کاملSubstitution of dietary oleic acid for myristic acid increases the tissue storage of α-linolenic acid and the concentration of docosahexaenoic acid in the brain, red blood cells and plasma in the rat.
Various strategies have been developed to increase the cellular level of (n-3) polyunsaturated fatty acids in animals and humans. In the present study, we investigated the effect of dietary myristic acid, which represents 9% to 12% of fatty acids in milk fat, on the storage of α-linolenic acid and its conversion to highly unsaturated (n-3) fatty acid derivatives. Five isocaloric diets were desi...
متن کاملMedium chain fatty acids as specific substrates for diglyceride acyltransferase in cultured hepatocytes.
The fatty acyl substrate specificity for de novo diglyceride, triglyceride, and phospholipid synthesis in cultured hepatocytes was evaluated. The fatty acyl availability for diglyceride and phospholipid synthesis is restricted to fatty acids longer than myristic acid with octanoic and decanoic acids being excluded. On the other hand, octanoic and decanoic acids may serve as substrates for the d...
متن کاملTissue-specific, nutritional, and developmental regulation of rat fatty acid elongases.
Of the six fatty acid elongase (Elovl) subtypes expressed in mammals, adult rat liver expresses four subtypes: Elovl-5 > Elovl-1 = Elovl-2 = Elovl-6. Overnight starvation and fish oil-enriched diets repressed hepatic elongase activity in livers of adult male rats. Diet-induced changes in elongase activity correlate with Elovl-5 and Elovl-6 mRNA abundance. Adult rats fed the peroxisome prolifera...
