Inhibition of thyroid hormone transport in plasma of rats by polychlorinated biphenyls.

چکیده
In this study the mechanism of thyroid hormone reductions reported in rodents, following exposure to TCDD (Potter et al, 1983), PCBs (GrassIe and Biesmann 1982; Brouwer 1987) and polybrominated biphenyls (Gupta et al 1983), was investigated in rats at the level of the transport protein, transthyretin (TTR), using 3,4,3',4'-tetrachlorobiphenyl (TCB) as a model compound. Moreover, the effeets of a commercial mixture of PCBs, Aroclor 1254, on plasma thyroid hormone concentrations was investigated in rats, in order to predict whether similar alterations would possibly occur in wildlife species, following environmental exposure to PCBs.

Placental transfer of a hydroxylated polychlorinated biphenyl and effects on fetal and maternal thyroid hormone homeostasis in the rat.
Earlier studies at our laboratory indicated that several hydroxylated polychlorinated biphenyls (OH-PCBs) detected in human blood could specifically inhibit thyroxine (T(4)) transport by competitive binding to the thyroid hormone transport protein transthyretin (TTR) in vitro. In the present study we investigated the effects of prenatal exposure to 5 mg/kg body weight of [14C]-labeled or unlabe...
متن کاملMaternal thyroid hormone increases HES expression in the fetal rat brain: an effect mimicked by exposure to a mixture of polychlorinated biphenyls (PCBs).
Thyroid hormone is known to be essential for normal brain development both before and after birth, but much less is known about the role of thyroid hormone development before birth. In rodents, thyroid hormone of maternal origin can selectively regulate gene expression in the fetal cortex; HES1 was identified as a putative thyroid hormone responsive gene in the fetal cortex. Using in situ hybri...
متن کاملPotential mechanisms of thyroid disruption in humans: interaction of organochlorine compounds with thyroid receptor, transthyretin, and thyroid-binding globulin.
Organochlorine compounds, particularly polychlorinated biphenyls (PCBs), alter serum thyroid hormone levels in humans. Hydroxylated organochlorines have relatively high affinities for the serum transport protein transthyretin, but the ability of these compounds to interact with the human thyroid receptor is unknown. Using a baculovirus expression system in insect cells (Sf9 cells), we produced ...
متن کاملEffect of polychlorinated biphenyls (PCB) on metabolism of thyroid hormone in Wistar rats.
Recently, goitrogenic action of DDT and P, P,-DDT on avian thyroid have been reported by Jefferies and French1,2). Fregly et al.3) observed an increase in thyroid weight of male rats by chronic treatment with DDT. Since these reports are based on the histological evidence and the measurment of thyroid weight, the mechanism of the action of DDT and its analogues is not known at present. DDT and ...
متن کاملThermodynamics Study of Polychlorinated Biphenyls (PCBs) Passing through SWNT and Their Removal from Environment
Polychlorinated Biphenyls (PCBs) have been widely used to flame-retard products common in homes and theworkplace, and subsequently. Polychlorinated biphenyls (PCBs) typify a class of highly toxic, stable aromaticpollutants. PCBs are recently being scrutinized for potential environment damage in groundwater and in theatmosphere. The elimination of chemical pollutants from a contaminated environm...
