Early Detection of Transplant Rejection by In Vivo F MRI

چکیده
Introduction Diagnosis of transplant rejection requires endomyocardial biopsy and entails risks. Here, we validated a F MRI approach for the early detection of organ rejection by noninvasive visualization of the macrophage host response. We employed biochemically inert emulsified perfluorocarbons (PFCs), which were reported to be preferentially phagocytized by monocytes and macrophages. Isografts from C57BL/6 and allografts from C57BL/10A mice were heterotopically transplanted into C57BL/6 recipients. In this model, the transplanted heart undergoes predictable progressive rejection, leading to graft failure after two weeks.

The goals of post-transplant management are to manage and treat postoperative complications, and develop a balanced long-term immunotherapy regimen that minimizes infection and side effects but controls rejection. While modern immunosuppressant regimes have reduced rates of graft loss due to rejection, they impart major risks for infection, growth failure, metabolic complications, and malignanc...

Dendritic cells (DCs) are a heterogeneous family of professional APCs involved in priming adaptive immune responses. Donor DCs (direct pathway of allorecognition) and recipient DCs presenting processed donor major histocompatibility complex (MHC) as peptides (indirect pathway of allorecognition) participate actively in graft rejection by stimulating recipient T cell responses following organ tr...

Background & Amis: BK polyomavirus (BKV) is common infection of childhood that persists in kidney epithelium. BKV reactivation characterized by viruria occurs in 35% to 57% of renal allograft recipients. BK virus was detected in serum samples in 6% to 29% of these patients. BKV-associated nephropathy occurs in as many as 8% of renal allograft recipients and may lead to . This study was carri...
متن کاملNon-invasive contrast enhanced MRI tissue characterisation early and late after heart transplantation
Background Non-invasive MRI tissue characterisation after HTX could potentially replace the hazardous myocardial biopsy for tissue staging and detection of transplant rejection. Recently, late gadolinium contrast enhanced cardiac MRI (LGE-CMR) showed infarct-typical patterns already early after HTX. Additionally, infarct-atypical LGE patterns were also shown in general but neither morphological...
متن کاملMRI Investigation of Macrophages in Acute Cardiac Allograft Rejection after Heart Transplantation Wu et al: MRI Explores Macrophage Roles in Heart Rejection
Background—Current immunosuppressive therapy after heart transplantation either generally suppresses the recipient’s entire immune system or is mainly targeting T-lymphocytes. Monocytes/macrophages are recognized as a hallmark of acute allograft rejection, but the roles that they play are not well characterized in vivo, because the tools for accessing in-situ macrophage infiltration are lacking...
