چکیده

Calcitonin receptors have been characterized for the first time in isolated osteoclasts. These receptors have been demonstrated by autoradiographic and biochemical methods, and the cells have also been shown to respond to calcitonin with a dose-dependent increase in cyclic AMP. The receptors in rat osteoclasts are specific and of high affinity (dissociation constant, Kd, 1 to 6 X 10-10 M), and are present in greater numbers than in any cell previously studied (>10' per cell). Chemical cross-linking of "SIlabeled salmon calcitonin to osteoclasts using disuccinimidyl suberate resulted in identification of a receptor component with a relative molecular weight of 80,000-90,000. By counting grains in autoradiographic experiments, we found that >80% of specifically bound radioactivity was associated with multinucleate osteoclasts and the remainder was associated with mononuclear cells that are not osteoblasts, but that may be osteoclast pre-

Biomaterial particle phagocytosis by bone-resorbing osteoclasts.

Abundant implant-derived biomaterial wear particles are generated in aseptic loosening and are deposited in periprosthetic tissues in which they are phagocytosed by mononuclear and multinucleated macrophage-like cells. It has been stated that the multinucleated cells which contain wear particles are not bone-resorbing osteoclasts. To investigate the validity of this claim we isolated human oste...

Intrathecal Amylin and Salmon Calcitonin Affect Formalin Induced c-Fos Expression in the Spinal Cord of Rats

Background: Amylin and Salmon Calcitonin belong to the calcitonin family of peptides and have high affinity binding sites in the rat spinal cord. The aim of this study was to characterize receptors for Amylin and Salmon Calcitonin functionally in the spinal cord of rats. We assessed the expression of c-Fos in response to intraplantar formalin in the lumbar regions of the spinal cord in consciou...

MECHANISM OF ACTION 1. Role of c-AMP: Calcitonin binds to specific calcitonin receptors on the plasma membrane of bone osteoclasts and renal tubular epithelial cells, activates adenyl cyclase

1. Role of c-AMP: Calcitonin binds to specific calcitonin receptors on the plasma membrane of bone osteoclasts and renal tubular epithelial cells, activates adenyl cyclase which increase c-AMP level which mediates the cellular effect of the hormone. This is the principal mechanism by which calcitonin acts. 2. Celluar Shift: It has been suggested that calcitonin may directly affect the relative ...

Osteoblasts release osteoclasts from calcitonin-induced quiescence.

There is an increasing body of indirect evidence that suggests that the localization and activity of osteoclasts, the major agents of bone resorption, are controlled by osteoblasts. In this paper I provide direct evidence that osteoblasts are indeed able to alter the behaviour of osteoclasts. I used calcitonin (CT) to suppress the cytoplasmic activity of osteoclasts isolated on glass or plastic...

EXTENDED REPORT Fibroblasts from the inner granulation tissue of the pseudocapsule in hips at revision arthroplasty induce osteoclast differentiation, as do stromal cells

Background: It has previously been shown that many osteoclast precursors are included in the granulation tissue within the pseudocapsule obtained at revision arthroplasty from hips with osteolysis. In vitro culture of only cells isolated from the granulation tissue has been previously shown to generate many mature osteoclasts. Objective: To investigate the presence or otherwise of supporting ce...
