AGE RELATED LIPID OXIDATION IN HUMAN LENS MEMBRANES BY INFRARED SPECTROSCOPY. Borchman D. Department of Ophthalmology and Visual Sciences, University of Louisville, Louisville Kentucky, 40202, USA. To measure primaty and secondary products of lipid Purnose oxidation with age in human lens membranes. Lipid oxidation is believed to contribute to a number of senescent diseases including cataract. Secondary products of lipid oxidation, such as malondialdehyde, have been measured in the lens and are not a good indicator of lipid oxidation. hence primaly and secondary products were measured in this stidy. Methods Lipids were extracted from 1 to 85 year old human lenses using a monophasic extraction protocol. Extracted lipid was dried on a AgCl window for infrared spectroscopic analysis. Infrared bands due to lipid oxidation were quantified: trans double bonds, hydroperoxides, carbonyls. These bands were compared to infrared bands representative of total lipid: the symmetric and asymmetric phosphate bends, CH stretching bands, cis double bond CH stretching band. Results Primary products of lipid oxidation increased linearly from 1 to 85 years. The ratio of trans to cis double bonds increased 7 fold during this period while all orher products increased 3 fold relative to one mole of phospholipid. Conclusions Lipid oxidation is substantial and increases linearly with age in the human lens. These products would be expected to increase membrane permeability diminish cation pump activity, crosslink proteins, destroy DNA and contribute the to the senescent related increase in membrane rigidity. A threshold of lipid oxidation could be reached upon which cataracts develop.

Analysis of grain elements and identification of best genotypes for Fe and P in Afghan wheat landraces

This study was carried out with the aim of developing the methodology to determine elemental composition in wheat and identify the best germplasm for further research. Orphan and genetically diverse Afghan wheat landraces were chosen and EDXRF was used to measure the content of some of the elements to establish elemental composition in grains of 266 landraces using 10 reference lines. Four elem...

Estimation of Trace Element Concentrations in Anti-Epileptic Medicinal Plants by Using EDXRF

Trace elemental analysis was carried out in 15 anti-epileptic medicinal plants using Energy Dispersive X-Ray Fluorescence (EDXRF) technique, with EX-3600, EDXRF spectrometer. The quantitative analysis is carried out using the software nEXT. The concentrations of elements like P, S, Cl, K, Ca, Mn, Fe, Cu, Zn, Br, Se, Rb, and Sr were estimated. The presence of some of these trace elements is corr...

Analysis of the elemental concentrations in rat femur's head using energy dispersive X ray fluorescence spectrometry

EDXRF spectrometry is used normally to determine the concentration of the different elements in a sample with advantages of good sensitivity, multielemental capabilities, fast, non-destructive and because of this simple relation to the fundamental physics of atom-radiation interaction, make of EDXRF a highly attractive analytical technique. A major goal for research in biomedical sciences over ...

Elemental Analysis of Two Rupee Indian Coins by EDXRF Technique

Energy Dispersive X-Ray Fluorescence (EDXRF) technique has been used to study two Rupee Indian coins minted during the period 1993 to 2010 for their elemental concentrations. From the analysis of the coins, it is found that the coins are made of binary alloy and it was observed that from 2005 onwards, the composition of the coins has been changed from Cupro-Nickel to Ferritic Stainless Steel (F...
