Comparison of ELISA, direct electron microscopy and antibody capture technique for detection of rotavirus in human stools--an experience in a developing country.

Two hundred and fourteen human faecal specimens were examined for detecting the presence of rotavirus by direct electron microscopy, antibody capture electron microscopy and ELISA. Twenty nine samples were positive with one of the three methods. Thirtyeight percent were positive with all three methods, 72.4% with ELISA, 62% with antibody capture electron microscopy and 55% with direct electron microscopy. ELISA was found to be the most sensitive method in our experience (JPMA 37: 172, 1987).

A total of 115 stools were examined for Rotavirus, using direct electron microscopy (EM) and Rotazyme. The overall agreement was 88.7%. Of the negative results, there was 91.95% agreement. Rotazyme reactions of three-plus or more gave a 100% agreement with EM. The Rotazyme test is a useful diagnostic aid in laboratories not capable of performing EM.
