Riboflavin-mediated Photosensitization of Vinca Alkaloids Distorts Drug Sensitivity Assays1

چکیده
Poor reproducibility of cytotoxicity tests with Vinca alkaloids has fre quently been reported. A commonly presumed light sensitivity of the drugs could not be confirmed. However, we found that they are photo sensitized by riboflavin (vitamin li.i, an obligatory component of cell culture media. Light of wavelengths below 500 nm triggered rapid photoreactions of riboflavin with vinblastine, \ im risiine, and vindesine in aqueous solutions. The photoreactions altered the absorption spectra of these alkaloids and yielded degradation products that could be separated by TLC. In cell cultures, both immediate and persisting, riboflavin-mediated photoreactivity could be distinguished. They preclude reliable determina tions of sensitivity and resistance to Vinca alkaloids, a.s exemplified on chemosensitive and multidrug-resistant mouse ascites cells. In experi ments involving photosensitization, the 50% inhibitory concentration val ues of sensitive and resistant cells were overlapping and fluctuated in the ranges from 3 to 30 nM and 15 to 360 TIMvinblastine, respectively. Corresponding values from series of experiments protected from photo sensitization were 1.02 ±0.22 nM and 18.5 ±3.42 nM. Hence, riboflavinmediated photoreactions must be fully prevented in assays of cellular drug sensitivity. Procedures for eliminating immediate as well as persisting photoreactivity were established.

Riboflavin-mediated photosensitization of Vinca alkaloids distorts drug sensitivity assays.
Poor reproducibility of cytotoxicity tests with Vinca alkaloids has frequently been reported. A commonly presumed light sensitivity of the drugs could not be confirmed. However, we found that they are photosensitized by riboflavin (vitamin B2), an obligatory component of cell culture media. Light of wavelengths below 500 nm triggered rapid photoreactions of riboflavin with vinblastine, vincrist...
متن کاملDNA damage increases sensitivity to vinca alkaloids and decreases sensitivity to taxanes through p53-dependent repression of microtubule-associated protein 4.
Taxanes and Vinca alkaloids are among the most active classes of drugs in the treatment of cancer. Yet, fewer than 50% of previously untreated patients respond, and clinicians have few ways of predicting who will benefit from treatment and who will not. Mutations in p53 occur in more than half of human malignancies and may alter the sensitivity to a variety of anticancer therapies. We have show...

Catharanthus roseus (L.) G.Don is a plant which produces anticancer and anti-mitotic indole alkaloids. Colchicine is an anti-mitotic drug. Anti-mitotic effects of Vinca herbacea Waldst. & Kit. indole alkaloids is unknown. The study were evaluated the antimitotic effect of alkaloids of V. herbacea, Catharanthus roseus andcolchicineon mitosis and microtubule arrangement of human blood cells. In t...
متن کاملDrug Delivery Systems and Combination Therapy by Using Vinca Alkaloids
Developing new methods for chemotherapy drug delivery has become a topic of great concern. Vinca alkaloids are among the most widely used chemotherapy reagents for tumor therapy; however, their side effects are particularly problematic for many medical doctors. To reduce the toxicity and enhance the therapeutic efficiency of vinca alkaloids, many researchers have developed strategies such as us...
متن کاملTubulin-targeted drug action: functional significance of class ii and class IVb beta-tubulin in vinca alkaloid sensitivity.
Aberrant expression of beta-tubulin isotypes is frequently described in tumor tissues and tubulin-binding agent (TBA)-resistant cell lines. There is limited understanding of the role of specific beta-tubulin isotypes in cellular sensitivity to TBAs, and to gain insights into the functional role of betaII- and betaIVb-tubulin, we examined these isotypes in lung cancer cell lines NCI-H460 (H460) ...
