چکیده

The control of cellular proliferation in mam malian tissues is currently being investigated in terms of the basic biochemical processes leading to deoxyribonucleic acid (DNA) synthesis and the means by which critical steps in the biosynthetic sequence may be regulated by product interaction. Various enzymatic reactions involved in the syn thesis of DNA precursor substances have been ex amined in growing and nongrowing tissues, and although some relationships have been observed which are consistent with growth demands there is yet no indication of a critical point at which control is initiated or a mechanism by which this is effected. Bucher (2) has recently reviewed much of the biochemical literature on these aspects of DNA synthesis in relation to liver regeneration. Some time ago we began to approach the problem of control at the level of deoxyribonucleoside tn

Nuclear and cytoplasmic pools of deoxyribonucleoside triphosphates in Chinese hamster ovary cells.

Nuclei were prepared from Chinese hamster ovary cells by fractionation in nonaqueous media. The distribution of the four common deoxyribonucleoside triphosphates between the nucleus and the cytoplasm was determined in cells during Gl and S phases and after inhibition of DNA synthesis by hydroxyurea or thymidine. During the Gl phase all triphosphates were present in both the cytoplasm and the nu...

Evaluation of ribonucleoside and deoxyribonucleoside triphosphate pools in cultured leukemia cells during exposure to methotrexate or methotrexate plus thymidine.

Continuous exposure to inhibitory concentrations of methotrexate produces distinct rates of steady-state growth of murine leukemia L1210 and human leukemia CCRF-CEM cells in culture. Addition of thymidine to the medium produces reversal (6 to 40%) of this steady-state growth rate inhibition. This study utilized combinations of methotrexate and thymidine for an evaluation of the accompanying rel...

Studies on bleomycin-induced repair DNA synthesis in permeable mouse ascites sarcoma cells.

To study the mechanism of DNA excision repair, a DNA repair system employing permeable mouse sarcoma (SR-C3H/He) cells was established and characterized. SR-C3H/He cells were permeabilized with a 0.0175% Triton X-100 solution. The permeable cells were treated with 1 mM ATP and 0.11 mM bleomycin, and then washed thoroughly to remove ATP and bleomycin. Repair DNA synthesis occurred in the bleomyc...

Specificity of Mouse Uridine Phosphorylase*

Two distinct pyrimidine nucleosidases, isolated from Escheri&a coli, show complete specificity for the pentose moiety of the nucleoside. Pyrimidine deoxyribonucleoside phosphorylase (1) shows complete specificity for the deoxyribosyl group, and uridine phosphorylase (2) for the ribosyl group. Mammalian uridine phosphorylase preparations (3, 4), however, have appreciable deoxyuridine phosphoryla...

Feedback Inhibition of Thymidine Kinase by Thymidine Triphosphate.

That the regulation of cell division and deoxyribonucleic acid synthesis is subject to some kind of chemical control in higher organisms is widely accepted (2-5). Davidson (2) has suggested that deoxyribonucleic acid synthesis takes place at critical concentrations of the deoxyribonucleoside triphosphates and that these compounds are components of a homeostatic mechanism. There are now at least...
