چکیده

Onoue, Kaoru, (Kyushu University, Fukuoka, Japan), Masayasu Kitagawa, and Yuichi Yamamura. Chemical studies on cellular components of Bordetella pertussis. I. Purification and properties of agglutinogen. J. Bacteriol. 82:648-656. 1961.-A method is described for the preparation of the agglutinogen of Hemophilus pertussis in phase I by methanol precipitation and ion-exchange chromatography. High homogeneity of this agglutinogen was evidenced by chromatography, but a trace of impurity was detected by ultracentrifugal analysis and by an agar gel diffusion test. The agglutinogen produced an allergic skin reaction in rabbits which had been immunized with whole phase I organisms. Chemical studies indicate that the agglutinogen should be a simple protein in nature. The approximate molecular weight was determined to be 10,000 by Archibald's method.

Proliferate Resonse to Purified Bordetella pertussis Toxin on Murine Spleen Lymphocytes

A purification procedure of pertussis toxin (PT) from submerged culture of Bordetella pertussis (B.pertussis) strain 134 using adsorption and affinity chromatography was discussed. The yield of the resulting PT was approximately 37.5mg/l of concentrated culture supernatant. The polypeptide pattern of the purified PT was investigated by SDS-PAGE and showed five bands between 11 to 26 KDa using l...

Antigens of Bordetella pertussis V. Separation of Agglutinogen 1 and Mouse-Protective Antigen.

Agglutinogen 1 of Bordetella pertussis strain 353/Z (serotype 1) was separated from protective antigen and histamine-sensitizing factor by starch-block electrophoresis. Most of the agglutinogen 1 migrated towards the cathode in starch-block electrophoresis, although some remained near the origin. Fractions containing most of the agglutinogen 1 were free of detectable mouse-protecting or histami...

Comparison of type 2 and type 6 fimbriae of Bordetella pertussis by using agglutinating monoclonal antibodies.

Two types of fimbriae have been identified on the pathogenic gram-negative organism Bordetella pertussis. Monoclonal antibodies to these fimbriae were produced to better understand the role of fimbriae as serotype-specific agglutinogens and to investigate the antigenic relationship between these fimbriae. Three monoclonal antibodies were identified that specifically agglutinated B. pertussis ce...

Virulence Factors Variation Among Bordetella Pertussis Isolates in Iran

Pertussis is still endemic and the recently resurgence of the disease caused by Bordetella pertussis has been shown in many countries. The polymorphism of the virulence genes of B. pertussis and lack of any information about the allelic variation between the Iranian isolates promotes us to analysis of the genes encoded virulence factors including ptxS1, prn, fim3 and cya to understand the diffe...

Purification and characterization of fimbriae isolated from Bordetella pertussis.

Fimbriae were detached from Bordetella pertussis by mechanical shearing and purified by successive precipitations with ammonium sulfate, phosphate buffer (pH 6.0), and magnesium chloride. In each of these purification steps, the fimbriae aggregated into bundles as seen by electron microscopy. These aggregates could be disaggregated at pH 9.5. By electron microscopy, the purified fimbriae appear...
