Glutaraldehyde nonfixation of isolated viral and yeast RNAs.

چکیده

The RNAs of brome mosaic (BMV), barley stripe mosaic (BSMV), and tobacco mosaic (TMV) viruses were inactivated by reaction with buffered glutaraldehyde. Glutaraldehyde did not fix 4% BMV-RNA, 20% t-RNA, 5% polyadenylic acid, or 5% adenosine monophosphate into water-insoluble precipitates, or gels, in distilled water or in low or high ionic strength buffers nor did it change their ultraviolet (UV) spectra. Two SDS- and phenol-purified commercial yeast RNA preparations from different sources gave UV spectra typical of pure RNA, but could not be freed of a contaminant that reacted with glutaraldehyde by forming a precipitate. The yeast RNAs did not become water-insoluble after glutaraldehyde reaction. BMV-RNA precipitated by Mg2+ could not be cross-linked into an insoluble form by glutaraldehyde. Nonfixation of RNA by glutaraldehyde must be considered in interpretation of attempts to localize RNA by electron microscopy.

Suppression of viral RNA recombination by a host exoribonuclease.

RNA viruses of humans, animals, and plants evolve rapidly due to mutations and RNA recombination. A previous genome-wide screen in Saccharomyces cerevisiae, a model host, identified five host genes, including XRN1, encoding a 5'-3' exoribonuclease, whose absence led to an approximately 10- to 50-fold enhancement of RNA recombination in Tomato bushy stunt virus (E. Serviene, N. Shapka, C. P. Che...

Isolation and Identification of Yeast Strains Capable of Producing Single Cell Protein from Whey in Co-Cultures with Saccharomyces cerevisiae

In this study, twenty-five whey samples collected from dairy industries in the city of Isfahan. The sampleswere cultured on malt extract broth (MEB) and yeast extract glucose chloramphenicol agar (YGCA) media.Eleven yeast strains (designated M1 to M11) were isolated from the culture. The strains were identified bytheir morphological and physiological properties. Betagalactosid...

Viral RNAs associated with ribosomes in Sindbis virus-infected HeLa cells.

Virus specific RNA ribosome complexes were isolated by sucrose density gradient centrifugation of cytoplasmic extracts from HeLa cells infected at 42 C with an RNA(+) mutant (ts2) of Sindbis virus. Viral RNA-ribosome complexes were accumulated by infected cells treated with sodium fluoride and cycloheximide. The RNA-ribosome complexes were characterized by (i) their sensitivity to the action of...

Biological Pretreatment of a Beverage Waste Using Yeast Isolated from the Factory Sludge (RESEARCH NOTE)

The beverage industry has been a heavy producer of organic pollution. The major contaminants found in waste are biodegradable organic compounds, volatile organic compounds, toxic metals, and recalcitrant xenobiotics, suspend solids, nutrients (Nitrogen and Phosphor) Microbial pathogens and parasites. Activated sludge flocs contain a wide range of microorganisms such as bacteria, Fungi, yeast, v...
