چکیده

The role of direct methods in macromolecular crystallography is discussed. The common belief that such methods will still remain marginal is rejected. Different sectors are analyzed. A direct procedure for phasing reflections when diffraction data of one isomorphous derivative are available is briefly described. The applications to experimental data of some test structures succeeded, and suggest that direct methods are competitive with traditional SIR techniques. Attention is also devoted to a formula which is able to recover the total from a partial structure. Direct methods can play a central role also for expanding (and refining) phases from derivative to native resolution, and can constitute an alternative to traditional molecular replacement techniques.

Liking likelihood.

Maximum-likelihood methods have now been applied to most areas of macromolecular crystallography, including data reduction, molecular replacement, experimental phasing and refinement. However, students of macromolecular crystallography are predominantly taught only traditional crystallographic methods and therefore have little understanding of the methods underlying the modern software that the...

Advances in spectroscopic methods for biological crystals. 2. Raman spectroscopy

A Raman microspectrophotometer is described that allows the spectroscopic investigation of protein crystals under exactly the same conditions as those used for X-ray data collection. The concept is based on the integration of the Raman excitation/collection optics into a microspectrophotometer built around a singleaxis diffractometer and a cooling device. It is shown that Raman spectra of outst...

The Interactive Relationship between Regional Trade Integration and Foreign Direct Investment

E conomic integration among countries has continued to deepen over the past decade. This is especially visible at the regional level, with the escalation of Regional Integration Agreements (RIAs) ranging from Free Trade Areas (FTAs) to Customs Unions (CUs). Nowadays, many developing countries have entered a new regional integration agreement with developed and developing countries. S...

The Macromolecular Crystallographic Information File (mmCIF)

Introduction The Protein Data Bank (PDB) format provides a standard representation for macromolecular structure data derived from X-ray diffraction and NMR studies. This representation has served the community well since its inception in the 1970's (Bernstein et al. 1) and a large amount of software that uses this representation has been written. However, it is widely recognized that the curren...
