Effect of methylthioinosine on nucleotide concentrations in L5178Y cells.

چکیده
phosphate concentrations declined, indicating turnover of this compound. Incubation of LSI78Y cells in medium containing 0.5 ^M MMPR reduced intracellular concentrations of adenosine triphosphate, guanosine triphosphate, and adenosine diphosphate by about 70% and proliferation rates by about 30%. Upon transfer of such cells to M MPR-free medium, nucleotide levels and proliferation rates returned to control values. L5178Y cells incubated in medium containing 0.1 ÕÕM MMPR proliferated at control rates even though their purine ribonucleotide levels were reduced by as much as 40%. The reduction in cellular pools of purine nucleotides during culture in 0.1 J/M MMPR was accompanied by an increase in the pyrimidine ribonucleotide concentrations. Cellular concentrations of uridine triphosphate and cytidine triphosphate increased almost threefold during the first 2 hr of exposure to 0. l ¿/M MPR, then fell during the next 2 hr to a steady-state level of about 180% of control values.

متن کاملQuantitation of intracellular metabolites of [35S]-6-mercaptopurine in L5178Y cells grown in time-course incubates.
6-Mercaptopurine (6MP) metabolism was quantitatively determined in L5178Y murine lymphoma. Cells grown in time-course incubates with [35S]-6MP were extracted with cold perchloric acid, and the buffered extracts were subjected to high-performance liquid cation-exchange chromatography prior to and after hydrolysis with alkaline phosphatase. Free sulfate, 6-thiouric acid, 6-thioxanthosine, 6-thiog...
متن کاملSynergistic effect of 6-mercaptopurine and 6-methylmercaptopurine ribonucleoside on the levels of adenine nucleotides of Sarcoma 180 cells.
The synergistic antitumor action of 6-mercaptopurine (6-MP) and 6-methylmercaptopurine ribonucleoside (MMPR) is confirmed with Sarcoma 180 ascites cells. Striking decreases are seen in the adenine nucleotide concentrations in cells incubated for 1 hr with this drug combination, and lesser effects are seen on the guanine nucleotide concentrations. Still greater decreases in adenine nucleotide le...
