چکیده

Estrogen has been implicated as a causal factor for anterior cruciate ligament injuries in women. Studies have demonstrated a decrease in anterior cruciate ligament fibroblast proliferation and collagen synthesis at supraphysiologic levels of estrogen in a rabbit model. Hypothesis: The authors hypothesized that physiologic levels of estrogen would have no significant effect on anterior cruciate ligament fibroblast proliferation and collagen synthesis in an ovine model. Methods: Anterior cruciate ligament fibroblasts were isolated from sheep knees using routine cell culture methods. The cells were exposed to 17β-estradiol at physiologic concentrations of 2.2, 5, 15, 25, 250, and 2500 pg/ml. Cell proliferation was determined by cell counts on days 4 and 6. Collagen synthesis was determined by H-proline incorporation on day 4. Immunohistochemistry was performed to detect estrogen receptors. Results: Immunohistochemistry demonstrated the presence of estrogen receptors in ovine anterior cruciate ligament fibroblasts. There was no significant difference in anterior cruciate ligament fibroblast proliferation or collagen synthesis regardless of 17βestradiol concentration. Conclusions: Based on results of this study, and given the low turnover of collagen in ligaments, it is unlikely that a 2to 3-day per month increase in circulating estrogen would result in rapid, clinically significant alterations in material properties of the anterior cruciate ligament in vivo. The etiology of noncontact anterior cruciate ligament injuries is complex and multifactorial in nature, meriting further investigation.

GDF-5 and bFGF Activate Matrix Synthesis and Integrin 2-mediated Cell Migration in Ligament Cells

INTRODUCTION: Anterior cruciate ligament (ACL) and medial collateral ligament (MCL) are the major stabilizers of knee joint. The deficiency of these ligaments could result in the early development of osteoarthritis. The cellular activities responding to growth factors have important roles in ligament healings. ACL has a poor healing potential rather than MCL after injuries. To accelerate the li...

GDF-5/7 and bFGF activate integrin alpha2-mediated cellular migration in rabbit ligament fibroblasts.

Cellular activities responding to growth factors are important in ligament healing. The anterior cruciate ligament (ACL) has poor healing potential compared to the medial collateral ligament (MCL). To assess the differences, we investigated the proliferation, migration, adhesion, and matrix synthesis responding to growth factors in rabbit ACL and MCL fibroblasts. ACL cell proliferation to basic...

Reduction of intraarticular adhesion of knee by local application of rapamycin in rabbits via inhibition of fibroblast proliferation and collagen synthesis

BACKGROUND The formation of intraarticular adhesion is a common complication after total knee arthroplasty or anterior cruciate ligament reconstruction. Previously, little research was reported regarding whether the local application of rapamycin (RAPA) could reduce intraarticular adhesion following knee surgery. In our present study, we determined the therapeutic effect of RAPA by local applic...

The mystery of female connective tissue.

FEMALES AND MALES DIFFER in many ways, including physiologically, and the fact that some types of connective tissue injuries (e.g., ligament ruptures and bone stress fractures) are more frequent in females than in males remains a puzzle. Estrogen receptors are known to be present in fibroblasts of tendon and ligaments. However, the effect of estrogen on anterior cruciate ligament collagen turno...

Mechanical Loading Improves Tendon-Bone Healing in a Rabbit Anterior Cruciate Ligament Reconstruction Model by Promoting Proliferation and Matrix Formation of Mesenchymal Stem Cells and Tendon Cells.

BACKGROUND/AIMS This study investigated the effect of mechanical stress on tendon-bone healing in a rabbit anterior cruciate ligament (ACL) reconstruction model as well as cell proliferation and matrix formation in co-culture of bone-marrow mesenchymal stem cells (BMSCs) and tendon cells (TCs). METHODS The effect of continuous passive motion (CPM) therapy on tendon-bone healing in a rabbit AC...
