Cross-sectional myofibre and myofibril growth in immobilized developing skeletal muscle.

چکیده
The effect of immobilization on skeletal muscle fibre and myofibril growth in the developing chick latissimus dorsi posterior muscle was investigated. Immobilization, immediately upon hatching, results in a 25 % decrease in mean myofibre cross-sectional area within 24 h. Longer periods of immobilization further retard normal myofibre growth until by day 27 after hatching the mean myofibre cross-sectional area was 70 % less than normal. The variation in individual myofibre cross-sectional areas (measured as the coefficient of variation, CV = standard deviation/mean x 100) was greater in immobilized muscles. The larger immobilized myofibres showed atypical myofibril and sarcotubular complex organization at the ultrastructural level of observation whereas the smaller immobilized myofibres appeared normal, except for their size. In 1to 27-day-old chicks, freeing the immobilized muscles for 24 h resulted in myofibril reorganization and sarcotubular proliferation but little myofibre growth. Freeing the immobilized muscles for 72 h resulted in a nearly complete recovery, in chicks up to 18 days post-hatching, of both cross-sectional myofibre area and ultrastrucrural morphology. Myofibres immobilized for longer than 18 days and subsequently freed for 72 h develop a normal myofibrillar and sarcotubular morphology but remain 23 to 29 % smaller in crosssectional area than myofibres from control muscles. Normal myofibril growth and organization are severely retarded in the immobilized myofibres. The evidence presented here suggests that sarcomere shortening causes growing myofibrils to rupture. The ruptured myofibrils are filled with proliferating elements of the sarcotubular complex which, in turn, completely subdivide (split) the myofibril.

Effects of disuse on growing and adult chick skeletal muscle.
The effects of long-term muscle inactivity, throughout post-hatching development, have been examined. Continuous immobilization of the chicken posterior latissimus dorsi (PLD) muscle from the first hour after hatching for varying periods up to 330 days, resulted in a significantly greater decrease in myofibre size (40-64% less than control) than occurred when adult muscles were immobilized for ...
متن کاملComparison of different skeletal muscles from growing domestic pigs and wild boars
The domestication process of pigs was associated with substantial morphological and physiological changes. The aim of this study was to comparatively investigate muscle microstructure and biochemical properties of four different skeletal muscles (M. longissimus, M. semitendinosus, M. psoas major, M. rhomboideus) in growing domestic pigs (n=5) and wild boars (n=4). The superiority of domestic pi...
متن کاملInsulin-like growth factor I slows the rate of denervation induced skeletal muscle atrophy.
Loss of the nerve supply to skeletal muscle results in a relentless loss of muscle mass (atrophy) over time. The ability of insulin-like growth factor-1 to reduce atrophy resulting from denervation was examined after transection of the sciatic nerve in transgenic MLC/mIGF-1 mice that over-express mIGF-1 specifically in differentiated myofibres. The cross sectional area (CSA) of all types of myo...
متن کاملSkeletal muscle atrophy occurs slowly and selectively during prolonged aestivation in Cyclorana alboguttata (Gunther 1867).
We investigated the effect of prolonged immobilisation of six and nine months duration on the morphology and antioxidant biochemistry of skeletal muscles in the amphibian aestivator Cyclorana alboguttata. We hypothesised that, in the event of atrophy occurring during aestivation, larger jumping muscles were more likely to be preserved over smaller non-jumping muscles. Whole muscle mass (g), mus...
متن کاملAn Electron Microscope Study of Myofibril Formation in Embryonic Chick Skeletal Muscle
The formation of myofibrils in the developing leg muscle of the 12-day chick embryo was studied by electron microscopy. Myofilaments of two varieties, thick (160-170 A in diameter) and thin (60-70 A in diameter), which have been designated myosin and actin filaments, respectively, on the basis of their similarity to natural and synthetic myosin and actin filaments, appear in the cytoplasm of de...
