چکیده

Electrochemotherapy is a new approach in the treatment of tumors that takes advantage of the permeabilization of the cell membrane by electric pulses to facilitate the delivery of chemotherapeutic drugs into the cells, According to the procedures described previously, the anti-tumor effectiveness of electrochemotherapy with bleomycin (BLM) was tested on three different murine tumor models, with different biological characteristics, to determine the variability in anti-tumor response. An arrest of growth of fibrosarcoma SA-I, malignant melanoma B-16 and Ehrlich ascites tumor (EAT) was observed in all mice subjected to electrochemotherapy, whereas neither BLM nor electric pulses had an effect on tumor growth when compared with controls. Partial and complete responses were also observed. The best anti-tumor response was observed for the SA-1 tumor model, where tumor growth delay was 31 days and 62% of the animals were free of tumor 100 days after the treatment. Side-effects of electrochemotherapy were demonstrated by body weight loss of the treated animals as well as some animal mortality recorded up to 7 days after the treatment, especially in the animals where tumors were located close to the spine. Our results are in accordance with previous results, and prove that use of high voltage electric pulses is promising for potentiation of BLM anti-tumor effectiveness.

Anti-tumor effectiveness of electrochemotherapy with bleomycin is increased by TNF-alpha on SA-1 tumors in mice.

With the aim to increase anti-tumor effectiveness of electrochemotherapy, adjuvant immunotherapy with tumor necrosis factor-alpha (TNF-alpha) was tested on tumors in mice. Increased anti-tumor effectiveness on SA-1 tumors was observed after combining TNF-alpha, injected either intratumorally or peritumorally, with electrochemotherapy using suboptimal dose of bleomycin (BLM). The increased anti-...

Tumor blood flow modifying effect of electrochemotherapy with bleomycin.

BACKGROUND Electrochemotherapy combines administration of the chemotherapeutic drug, followed by application of electric pulses in order to increase drug delivery into the cells. The aim of this study was to determine the tumor blood flow modifying effect of electrochemotherapy with bleomycin and correlate it with its antitumor effectiveness and extent of tumor necrosis. MATERIALS AND METHODS...

Intratumoral cisplatin administration in electrochemotherapy: antitumor effectiveness, sequence dependence and platinum content.

Electrochemotherapy using intratumoral cisplatin administration was tested on EAT tumors in mice. Mice were treated with eight electric pulses (100 micros, 1 Hz, 1040 V) and/or cisplatin (1, 2, 4 and 8 mg/kg). Cisplatin treatment resulted in up to 20 days of tumor growth delay. Electrochemotherapy resulted in tumor cures; local tumor control reached a plateau at 4 mg/kg in 67% of tumor cures. T...

Electrochemotherapy with cisplatin: clinical experience in malignant melanoma patients.

Electrochemotherapy consists of chemotherapy followed by local application of electric pulses to the tumor to increase drug delivery into the cells. The aim of this Phase II clinical study was to evaluate the antitumor effectiveness of electrochemotherapy using intratumoral cisplatin administration on cutaneous tumor nodules in malignant melanoma patients. In 10 patients, 133 tumor nodules of d...

Treatment of Murine Tumor Models of Breast Adenocarcinoma by Continuous Dual-Frequency Ultrasound

Introduction: Acoustic transient cavitation is the primary mechanism of sonochemical reaction and has potential use for tumor treatment. In this study, the in vivo anti-tumor effect of simultaneous dual-frequency ultrasound at low-level intensity (ISATA < 6 W/cm2) was investigated in a spontaneous murine model of breast adenocarcinoma in Balb/c mice. Materials and Methods: Forty tumor bearing m...
