Visualizing Bacterial Cell Walls and Biofilms
نویسنده
چکیده
T he surfaces of microbial cells are perhaps their most important structure because they are in immediate contact with the external environment. Microbial cell walls admit nutrients and release wastes, and yet they must resist internal turgor pressure and environmental insults and maintain cellular shape. Walls also help cells adhere to other surfaces and provide space for specialized structures such as flagella, pili, spinae, capsules, S-layers, and exopolymeric substances (EPS). Delving into the native architecture of gramnegative and gram-positive bacterial surfaces traditionally depended on electron microscopy, which uses high vacuums and high electron accelerating voltages. These requirements usually ensure that only dry specimens can be imaged, yet living cells and their structures rely on water and its astounding ability to interact with and configure molecular ingredients. However, advances in cryo-transmission electron microscopy (cryoTEM) are helping to overcome this difficulty because they allow investigators to examine native, hydrated structures in bacteria and their associated biofilms.

In Vitro Binding Potentials of Bentonite, Yeast Cell Wall and Lactic Acid Bacteria for Aflatoxin B1 and Ochratoxin A
Background: This study intended to assess individual and combined adsorption potentials of three adsorbents (processed bentonite as an inorganic adsorbent, and cell walls of Saccharomyces cerevisiae and of the GG strain of Lactobacillus rhamnosus as organic adsorbents) for aflatoxin B1 and ochratoxin A under in vitro conditions. Methods: This study was conducted in Ferdowsi University of Mashh...
متن کاملStenosis triggers spread of helical Pseudomonas biofilms in cylindrical flow systems
Biofilms are multicellular bacterial structures that adhere to surfaces and often endow the bacterial population with tolerance to antibiotics and other environmental insults. Biofilms frequently colonize the tubing of medical devices through mechanisms that are poorly understood. Here we studied the helicoidal spread of Pseudomonas putida biofilms through cylindrical conduits of varied diamete...
متن کاملEffects of biofilm formation in bacteria from different perspectives
Bacterial communities are able to form complex and three-dimensional biofilm structures. Biofilm formation is an ancient and integral component of the prokaryotic life cycle and a key factor for survival in diverse niches. In biofilms, bacterial lifestyle changes from free-floating cells to sessile cells. Presence in biofilms gives new traits to bacteria, which distinguish them from free cells....
متن کاملCalcium binding to S. mutans grown in the presence or absence of sucrose.
Sucrose is the most cariogenic dietary carbohydrate because it is a substrate for insoluble extracellular polysaccharide (IEPS) production in dental biofilms, which can proportionally decrease bacterial density and, consequently, the number of biofilm calcium (Ca) binding sites. Ca bound to bacterial cell walls can be released into the biofilm fluid during a cariogenic challenge, reducing the d...
متن کاملA mechanistic study on the antibacterial behavior of silver doped bioceramic
Antibacterial bioceramics were successfully fabricated by adding 0.5 wt% silver to yttria-stabilized zirconia (YSZ) and their surface antibacterial properties were investigated. Silver-containing YSZ bioceramics exhibited superior antibacterial rate against both Staphylococcus aureus (99.9%) and Escherichia coli cells (88.6%) compared with non-doped-YSZ. The antibacterial action of silver was s...
