Loss of G Protein 7 Alters Behavior and Reduces Striatal olf Level and cAMP Production

چکیده
The G protein -dimer is required for receptor interaction and effector regulation. However, previous approaches have not identified the physiologic roles of individual subtypes in these processes. We used a gene knockout approach to demonstrate a unique role for the G protein 7-subunit in mice. Notably, deletion of Gng7 caused behavioral changes that were associated with reductions in the olf-subunit content and adenylyl cyclase activity of the striatum. These data demonstrate that an individual -subunit contributes to the specificity of a given signaling pathway and controls the formation or stability of a particular G protein heterotrimer.

Up-regulation of AGS3 during morphine withdrawal promotes cAMP superactivation via adenylyl cyclase 5 and 7 in rat nucleus accumbens/striatal neurons.
Effective medical treatment of opiate addiction is limited by a high relapse rate in abstinent addicts. Opiate withdrawal causes cAMP superactivation, but the underlying molecular mechanisms are not clear. Recent evidence implicates an activator of G-protein signaling 3 (AGS3) in opiate addiction. We found previously that during a 10-min activation of opioid receptors, AGS3 binds G alpha(i)-GDP...
متن کاملGalpha(olf) levels are regulated by receptor usage and control dopamine and adenosine action in the striatum.
In the striatum, dopamine D(1) and adenosine A(2A) receptors stimulate the production of cAMP, which is involved in neuromodulation and long-lasting changes in gene expression and synaptic function. Positive coupling of receptors to adenylyl cyclase can be mediated through the ubiquitous GTP-binding protein Galpha(S) subunit or through the olfactory isoform, Galpha(olf), which predominates in t...

Although L-3,4-dihydroxyphenylalanine (L-DOPA) remains the reference treatment of Parkinson's disease, its long-term beneficial effects are hindered by L-DOPA-induced dyskinesia (LID). In the dopamine (DA)-denervated striatum, L-DOPA activates DA D₁ receptor(D₁R) signaling, including cAMP-dependent protein kinase A (PKA) and extracellular signal-regulated kinase (ERK), two responses associated ...
متن کاملEffect of Prenatal Stress on Offspring's Learning, Memory and Coping Strategies
Introduction: The prenatal stress is a key factor which affects the growth and function of the brain. Several studies have shown that prenatal stress induces deficits in learning and memory of the offspring. The prenatal stress alters the activity of neurotransmitters, such as noradrenaline, dopamine and serotonin, via over-activation of the hypothalamic–pituitary–adrenal axis. In addition, the...
متن کاملG(olf) and Gs in rat basal ganglia: possible involvement of G(olf) in the coupling of dopamine D1 receptor with adenylyl cyclase.
Using specific antibodies and cDNA probes, we have investigated, in rat basal ganglia, the distribution and the regulation of the expression of the alpha subunits of Gs and G(olf), two GTP-binding proteins (G-proteins) that stimulate adenylyl cyclase. We confirmed that G(olf) alpha is highly expressed in caudate-putamen, nucleus accumbens, and olfactory tubercle, whereas Gs alpha is less abunda...
