چکیده

Correlative light and electron microscopy (CLEM) is a useful tool for investigating cellular structures at high-resolution. We have extended this technology and developed a new method in which CLEM is combined with live-cell fluorescence imaging, “Live CLEM”. With this method, living cells are first examined by fluorescence microscopy then fixed in situ, and finally the same cells are subjected to electron microscopy. This simple yet powerful method combines temporal and high resolution spatial information. However, due to technical difficulties in immobilizing non-adherent target cells during sample preparation, this method has usually been applied to the study of adherent cells. Here we report a method of live CLEM that can be applied to yeast cells. Since yeast are a powerful tool in molecular genetics, application of this method to the study of yeast should greatly advance our understanding of the molecular basis of the function of cellular structures.

An Improved Procedure for Subcellular Spatial Alignment during Live-Cell CLEM

Live-cell correlative light and electron microscopy (CLEM) offers unique insights into the ultrastructure of dynamic cellular processes. A critical and technically challenging part of CLEM is the 3-dimensional relocation of the intracellular region of interest during sample processing. We have developed a simple CLEM procedure that uses toner particles from a laser printer as orientation marks....

Controlled Light Exposure Microscopy (CLEM): Photobleaching and Phototoxicity Reduced

Controlled Light Exposure Microscopy (CLEM): Photobleaching and Phototoxicity Reduced Photobleaching and phototoxicity are often the Achilles‘ heels of fluorescence live-cell imaging. Controlled Light Exposure Microscopy (CLEM) is a novel, simple imaging approach that reduces photobleaching and phototoxicity 2 to 10-fold without compromising image quality. The basic concept of CLEM is that ligh...

Topic Introduction Live Cell Imaging of Yeast

The development of cloning vectors for green fluorescent protein (GFP) and the simplicity of yeast reverse genetics allow straightforward labeling of yeast proteins in living cells. Budding and fission yeast are therefore attractive organisms in which to study dynamic cellular processes such as growth, cell division, and morphogenesis using live cell fluorescence microscopy. This article focuse...

D correlative light and electron microscopy of cultured cells using serial blockface scanning electron microscopy Matthew

The processes of life take place in multiple dimensions, but imaging these processes in even three dimensions is challenging. Here we describe a workflow for 3D correlative light and electron microscopy (CLEM) of cell monolayers using fluorescence microscopy to identify and follow biological events, combined with serial blockface scanning electron microscopy to analyse the underlying ultrastruc...

Correlative light and electron microscopy (CLEM) as a tool to visualize microinjected molecules and their eukaryotic sub-cellular targets.

The eukaryotic cell relies on complex, highly regulated, and functionally distinct membrane bound compartments that preserve a biochemical polarity necessary for proper cellular function. Understanding how the enzymes, proteins, and cytoskeletal components govern and maintain this biochemical segregation is therefore of paramount importance. The use of fluorescently tagged molecules to localize...
