Optimization of Protease Production by Bacillus mojavensis A21 on Chickpea and Faba Bean

چکیده
Response surface methodology (RSM) was employed to optimize the medium composition and culture conditions for the production of alkaline proteases by Bacillus mojavensis A21 on uncommon substrates: chickpea (CF) and faba bean (FF) flours. A significant positive influence of temperature, CF, FF, incubation time and inoculums size on the protease production was evaluated by Plackett Burman Design. Among these, CF was the most influential factor. The enhancement of protease to 9127 U/ml was achieved with the optimization procedure on the medium composed of (g/l): CF, 40; FF 30, NaCl 2.0; KH2PO4 1; K2HPO4 1; CaCl2, 0.1; MgSO4 0.1. The cultures were conducted for 72 hours with an IS of 2%, at 30 ̊C, an agitation speed of 150 rpm and an initial pH of 8.0. More interestingly, the optimization was accomplished using two cheap and local fermentation substrates, CF and FF, which could result in a significant reduction in the cost of medium constituents. The maximum alkaline protease production was 9127 U/ml after 72 h of incubation and showed 5-fold increase in protease production over the initial level.

Response of surface optimization for the enhanced production of alkaline protease isolated from Bacillus sp. with bean husk as a new substrate
Optimization of the fermentation medium for maximum alkaline protease production was carried out. Fifteen positive isolates were examined for their extent of alkaline protease production. The most potent producer was identified as Bacillus sp. The solid substrate screening showed that the combination of wheat straw and bean husk was the best one. The initial screening by using Plackett–Burman’s...

The New Crops Program of Virginia State University, established in 1991, has evaluated the production feasibility of a wide array of leguminous crops including chickpea, faba bean, mungbean, and pigeonpea under Virginia’s agro-climatic conditions (Bhardwaj et al. 1996). Such crops could provide alternatives to farmers in Virginia and adjoining Mid-Atlantic States. These farmers, in general, rel...

The production of protease by Bacillus licheniformis BBRC 100053 was studied. The most appropriate medium for the growth and protease production is composed of: lactose 1%, yeast extract 0.5%, peptone 0.5%, KH2PO4 0.1%, MgSO4.7H2O 0.02%. Enzyme production corresponded with growth and reached a maximums level (589 U/ml) during the stationary phase at 35°C, pH equivalent to 10 and with 150 rpm af...
