INTERACTION OF GOLD NANOPARTICLES WITH RAT BRAIN SYNAPTOSOMAL PLASMA MEMBRANE Na/K- ATPase AND Mg-ATPase

چکیده
The aim of the work was to investigate the interaction between borate capped gold nanoparticles (NPs) and the rat brain synaptosomal plasma membranes (SPM), as well as the effects of these NPs on SPM Na/KATPase and Mg-ATPase activity. The changes in the UV-vis spectra of NPs and SPM assembly suggested the agglomeration and precipitation of NPs. FTIR measurements indicated that both protein –SH and –NH2 groups and positively charged membrane fragments were implicated in the adhesion of SPM to the surface of NPs. AFM showed an increase in the particularization of the SPM material after mixing with gold NPs. Influence of gold NPs on Na/K-ATPase and MgATPase activity was investigated as the function of NPs and protein concentration, preincubation time and also in the presence of various concentrations of ouabain, the specific enzyme inhibitor. NPs induced the stimulation of Na/K-ATPase activity for more than 100%, since Mg-ATPase activity reminded unaffected. We propose that this stimulation of enzyme activity was a consequence of an increase of the active surface of membranes.

Sensitivity of the brain synaptosomal membrane Mg(2+)-ATPase activity to arachidonic acid is under control of the Na+,K(+)-ATPase state.
Evidence is presented for the sensitivity of the synaptosomal plasma membrane Mg(2+)-ATPase activity to arachidonic acid being dependent on the functional state of Na+,K(+)-ATPase. An "Inversion effect" was observed at arachidonic acid concentrations exceeding 80 mumol/l when the Mg(2+)-ATPase activity (after ouabain addition) is higher than the total ATPase activity (without ouabain). The "Inv...
متن کاملMechanisms of L-Triiodothyronine-Induced Inhibition of Synaptosomal Na+-K+-ATPase Activity in Young Adult Rat Brain Cerebral Cortex
The role of thyroid hormones (TH) in the normal functioning of adult mammalian brain is unclear. Our studies have identified synaptosomal Na(+)-K(+)-ATPase as a TH-responsive physiological parameter in adult rat cerebral cortex. L-triiodothyronine (T3) and L-thyroxine (T4) both inhibited Na(+)-K(+)-ATPase activity (but not Mg(2+)-ATPase activity) in similar dose-dependent fashions, while other ...
متن کاملProteolytic digestion of Cl(-)-ATPase in rat brain synaptosomes.
Upon tryptic digestion of synaptosomes, ATPase activities decreased in the order of Cl(-)-ATPase greater than or equal to Na+,K+-ATPase greater than anion-insensitive Mg2+-ATPase. Upon synaptosome treatment with hypotonic solution, Cl(-)-ATPase or anion-insensitive Mg2+-ATPase was slightly inactivated, while Na+,K+-ATPase underwent a much larger degree of inactivation. ATP-Mg inhibited the ATPa...
متن کاملO-10: A Marked Animal-Vegetal Polarity in The Localization of Na+,K+-ATPase Activity and Its Down-Regulation Following Progesterone-Induced Maturation
Background: Polarized cells are key to the process of differentiation. Xenopus oocyte is a polarized cell that has complete blue-print to differentiate 3 germ layers following fertilization, as key determinant molecules (Proteins and RNAs) are asymmetrically localized. The objective of this work was to localize Na+, K+-ATPase activity along animal-vegetal axis of polarized Xenopus oocyte and fo...
متن کاملEffect of chronic exposure to aluminium on isoform expression and activity of rat (Na+/K+)ATPase.
The ability of aluminum to inhibit the (Na(+)/K(+))ATPase activity has been observed by several investigators. The (Na(+)/K(+))ATPase is characterized by a complex molecular heterogeneity that results from the expression and differential association of multiple isoforms of both catalytic (alpha) and regulatory (beta) subunits. For instance, three main alpha (alpha(1), alpha(2) and alpha(3)) and...
