[Intravenous inoculation direct challenge method for potency test of Japanese encephalitis vaccine on suckling mice].

چکیده
The potency test of Japanese encephalitis (JE) vaccine is routinely carried out by intracerebral challenge of immunized mice using the Nakayama strain of JE virus, however since natural infection is by the peripheral route with a wild strain, one of the authors has reported (Bull. WHO, 43, 553, 1970) a new method by a subcutaneous challenge test on suckling mice. Since the usual route of infection is the result of mosquito bite, the authors developed a new challenge test by the intravenous route. Suckling mice 9-13 days old were immunized twice intraperitoneally with 0.15ml of different serial dilutions of vaccine on day 1 and 4. A challenge dose of JE virus equal to 500 icLD50 for suckling mice was administered into the tail vein on day 8, when the mice were not more than 20 days old. The JaGAr-01 strain was used for the challenge virus as it had been more recently isolated and is more virulent than the Nakayama strain. This method should give a more accurate assessment of the effect of vaccine the purpose of which is to protect humans from infection.

Comparative Assessment of the Whole-cell Pertussis Vaccine Potency Using Serological and Intracerebral Mouse Protection Methods
One of the most important QC tests of whole-cell pertussis vaccine (WCPV) is potency test. In this regard, mouse protection test (MPT) is the current potency method, which is associated with severe animal distress and large variability in results. The purpose of this study was to assess Pertussis Serological Potency Test (PSPT) as a serological alternative method to intracerebral challenge in M...

The protective ability of cytotoxic T cells (CTL) raised in vitro against Japanese encephalitis virus (JEV) was examined by adoptive transfer experiments. Adoptive transfer of anti-JEV effectors by intracerebral (i.c.) but not by intraperitoneal (i.p.) or intravenous (i.v.) routes protected adult BALB/c mice against lethal i.c. JEV challenge. In contrast to adult mice, adoptive transfer of anti...
متن کاملRecombinant sindbis/Venezuelan equine encephalitis virus is highly attenuated and immunogenic.
Venezuelan equine encephalitis virus (VEEV) is an important, naturally emerging zoonotic virus. VEEV was a significant human and equine pathogen for much of the past century, and recent outbreaks in Venezuela and Colombia (1995), with about 100,000 human cases, indicate that this virus still poses a serious public health threat. The live attenuated TC-83 vaccine strain of VEEV was developed in ...
متن کاملInfectivity of Plasmodium berghei sporozoites delivered by intravenous inoculation versus mosquito bite: implications for sporozoite vaccine trials.
Plasmodium berghei sporozoites delivered by mosquito bite were more infectious to outbred CD-1 mice than were sporozoites delivered by intravenous inoculation. The route of challenge also affected vaccine efficacy. In view of these findings and the fact that mosquito bites are the natural mode of sporozoite delivery, infectious mosquito bites should be considered the challenge protocol of choic...
متن کاملProtective mechanisms induced by a Japanese encephalitis virus DNA vaccine: requirement for antibody but not CD8(+) cytotoxic T-cell responses.
We have previously shown that a plasmid (pE) encoding the Japanese encephalitis virus (JEV) envelope (E) protein conferred a high level of protection against a lethal viral challenge. In the present study, we used adoptive transfer experiments and gene knockout mice to demonstrate that the DNA-induced E-specific antibody alone can confer protection in the absence of cytotoxic T-lymphocyte (CTL)...
