چکیده

The globus pallidus plays a central integrative role in the basal ganglia circuitry. Morphological studies have revealed a high level of GABA and GABA(A) receptors in the globus pallidus. To further investigate the effects of endogenous GABA(A) neurotransmission in the globus pallidus of normal and parkinsonian rats, in vivo extracellular recording and behavioral tests were performed in the present studies. In normal rats, micro-pressure ejection of GABA(A) receptor antagonist gabazine (0.1 mM) increased the spontaneous firing rate of pallidal neurons by 28.3%. Furthermore, in 6-hydroxydopamine parkinsonian rats, gabazine increased the firing rate by 46.0% on the lesioned side, which was significantly greater than that on the unlesioned side (21.5%, P < 0.05), as well as that in normal rats (P < 0.05). In the behaving rats, unilateral microinjection of gabazine (0.1 mM) evoked consistent contralateral rotation in normal rats, and significantly potentiated the number of apomorphine-induced contralateral rotations in parkinsonian rats. The present electrophysiological and behavioral findings may provide a rational for further investigations into the potential of pallidal endogenous GABA(A) neurotransmission in the treatment of Parkinson's disease.

The effect of intrastriatal injection of estrogen on pallidal field potential and rigidity in Parkinsonian -ovariectomized rats

The major pathological feature of Parkinson’s disease (PD) is a progressive loss of dopamine-producing neurons of the substantia nigra pars compacta (SNc), resulting in a reduction of dopamine (DA) content in the target field of these neurons, the striatum (STR). The present evidences suggest that female sex hormones may influence the onset and severity of PD symptoms. PD is more prevalent in m...

Effects of substance P on neuronal firing of pallidal neurons in parkinsonian rats.

Substance P is an important neurotransmitter or neuromodulator in central nervous system. Morphological studies have revealed the existence of substance P and its high affinity receptor, neurokinin-1 receptor, in globus pallidus. The expression of neurokinin-1 receptor in external globus pallidus has been reported to be decreased or unchanged in parkinsonian patients. To further investigate the...

Adenosine A2A Receptor Modulates the Activity of Globus Pallidus Neurons in Rats

The globus pallidus is a central nucleus in the basal ganglia motor control circuit. Morphological studies have revealed the expression of adenosine A2A receptors in the globus pallidus. To determine the modulation of adenosine A2A receptors on the activity of pallidal neurons in both normal and parkinsonian rats, in vivo electrophysiological and behavioral tests were performed in the present s...

Activity of pallidal and striatal tonically active neurons is correlated in mptp-treated monkeys but not in normal monkeys.

The goal of this study is to assess the function of tonically active neurons (TANs) of the striatum and their malfunction in the parkinsonian state. We recorded multiple spike trains of striatal TANs and pallidal neurons, which are the main target of striatal projections. Recordings were performed in two vervet monkeys before and after the induction of tremulous parkinsonism by systemic injecti...
