چکیده

DUXBURY, RALPH E., SADUN, ELVIO H., SCHOENBECHLER, MAURICE J., AND STROUPE, DAVID A. 1974. Tryparwsomu rhodesiense: Protection in mice by inoculations of homologous parasite products. Experimental Parasitology 36, 70-76. Mice were immunized against Trypanosoma rhodesiense (Wellcome strain) with whole lyuphilized trypanosomes, with antigens produced by disrupting lyophilized trypanosomes under pressure, and with excretions and s#ecretions of the living parasites. The survival rate in groups of 40 mice inoculated with ‘disrupted trypansomes and challenged with the homologous strain was 48% with a soluble fraction, and 70% with a particulate fraction of the parasites. There was 95% survival after challenge in a group immunized with lyophilized trypanosomes; none of the controls survived. Results were essentially the same whether or not an aluminum hydroxide adjuvant was used. In subsequent experiments, complete promotion was obtained with either crude excretion-secretion (ES) antigens or the particulate fraction of the ES antigen, while 40% of the mice survived challenge after inoculations of ES supernatant fluid. Mice immunized with crude ES antigen failed to survive challenge with a heterologous strain, although their mean survival time was prolonged several days beyond that of the controls. INDEX DESCRIPTORS: Trypanosomu ddesiense; Immunization; Antigens, excretion and secretion; Lyophilization; Pressure cell fractionation; Mice.

Immunization with a tubulin-rich preparation from Trypanosoma brucei confers broad protection against African trypanosomosis.

Tubulin from Trypanosoma brucei was purified to near homogeneity using a protocol which involved treatment with urea with subsequent renaturation and was then used to immunize mice. Renatured tubulin further purified by SDS-PAGE (denatured), synthetic tubulin peptides (STP), and rat brain tubulin (RbTub) were also used. Immunized mice were challenged with T. brucei, Trypanosoma congolense or Tr...

Trypanosoma rhodesiense infection in mice: sex dependence of resistance.

There is large variation in the survival of inbred mouse strains infected with Trypanosoma rhodesiense (EATRO 1886). Of those strains that survived for at least 22 days postinfection, female mice were markedly more resistant than male mice. The longer a strain survived, the greater was the difference in survival between male and female mice. Parasite counts were higher in male mice than in fema...

A flagellar pocket membrane fraction from Trypanosoma brucei rhodesiense: immunogold localization and nonvariant immunoprotection.

In contrast to the abundance of detailed information on variant-specific surface coat antigens of African trypanosomes, data on possible common or nonvariant antigens within these protozoa are surprisingly limited. In this study, the cellular localization and protective potential of a previously characterized flagellar pocket membrane (FPM) fraction were determined. Immunogold staining of live ...

Trypanosomiasis in mice with naturally occurring immunodeficiencies.

By using mice with naturally occurring defects, we have shown that an intact macrophage system is crucial to survival with the pathogenic protozoan Trypanosoma rhodesiense, since a defect in these cells decreased survival by half. Deficiencies in natural killer cell function or complement levels had no effect on survival. However, the capacity to survive trypanosomiasis was not related to the l...

Development of drug resistance in Trypanosoma brucei rhodesiense and Trypanosoma brucei gambiense. Treatment of human African trypanosomiasis with natural products (Review).

Human African trypanosomiasis is an infectious disease which has resulted in the deaths of thousands of people in Sub-Saharan Africa. Two subspecies of the protozoan parasite Trypanosoma brucei are the causative agents of the infection, whereby T. b. gambiense leads to chronic development of the disease and T. b. rhodesiense establishes an acute form, which is fatal within months or even weeks....
