Carotenoid composition of spinach chloroplast grana and stroma lamellae.

چکیده
Stroma lamellae and grana stacks prepared by French press rupture of spinach (Spinacia oleracea) chloroplasts contain similar amounts of beta-carotene on a protein basis. The grana fraction has considerably more xanthophylls than does the stroma fraction. Total carotenoid to chlorophyll ratios are similar for both fractions.

The site of ferricyanide photoreduction in the lamellae of isolated spinach chloroplasts: a cytochemical study.
Ferricyanide was used as a Hill oxidant to localize the site of photoreduction in chloroplast lamellae. The ferrocyanide formed on illumination was complexed with copper ions to form insoluble, electron-dense precipitates of Cu ferrocyanide, which can be easily seen in unstained preparations in the electron microscope. Control experiments showed no precipitates in the dark, at zero time, in the...
متن کاملBiosynthesis of grana and stroma lamellae in spinach.
Park and co-workers (Annu. Rev. Plant Physiol. 22: 395-430) have suggested that stroma lamellae, which perform only photosystem I contain a developing photosystem II which becomes functional upon the folding of these membranes to form grana stacks. The present study was undertaken to test this hypothesis. The lipid and protein constituents of chloroplast membranes were pulse-labeled radioactivel...
متن کاملThe association of chloroplast DNA with photosynthetic membrane vesicles from spinach chloroplasts.
To investigate the association between chloroplast DNA (cp DNA) and the photosynthetic membranes of spinach chloroplasts, previously suggested by electron-microscope autoradiography, use has been made of vesicles formed by isolating chloroplasts directly in 3.5 mM Mg2+. These chloroplast vesicles consist of photosynthetic membranes, separate from chloroplast envelope membranes. Light and electr...
متن کاملLocalization of Chlorophyll in Spinach Chloroplast Lamellae by Fluorescence Microscopy
Chlorophyll in green plants is thought to reside in the internal membranes or lamellae of chloroplasts (1). These membranes are observable by electron microscopy and can be roughly grouped, on the basis of size and arrangement , into grana lamellae and stroma lamellae which exist as closed, flattened sacs, or thylakoids (2). Al though the chlorophyll molecule is not yet demonstrable by electron...
متن کاملThe Effect of Manganese Deficiency on the Structure of Spinach Chloroplasts
Manganese is involved in the photosynthetic reactions of algae and leaves of higher plants (1-3). In tomato, manganese deficiency results in chloroplasts with a greatly reduced Hill reaction activity per unit chlorophyll (3). In spinach, manganese deficiency causes a 50 to 70 per cent reduction in the FMN-mediated photophosphorylation of isolated chloroplasts but only slightly affects the phosp...
