Intrinsically Disordered Proteins in Biomineralization

چکیده
Intrinsically disordered proteins (IDPs) have the potential to play a unique role in the study of proteins and the relationships between structure and function. Intrinsic disorder affects chemical and cellular events such as cell signaling, macromolecular self-assembly, protein removal and crystal nucleation and growth. This chapter explores the structural principles by which IDPs act and reveals the prevalence of IDPs in the field of biomineralization. It has been demonstrated that proteins involved in biomineralization are frequently very extended and disordered. Moreover, the disordered structure is integral to how these proteins fulfill their functions. We have focused on the analysis of polypeptide folding, the role of posttranslational modifications, predictions of the structural disorder and the degree of disorder in secondary structures. Computational and biophysical strategies to analyze the secondary structures and evaluate the degree and nature of "disorder" in proteins are described. Biomineralization is the result of the orchestration of a series of protein-protein, proteinmineral and protein-cell interactions. Identifying unfolded functional domains in cell signaling may have a great impact in the study of tissue regeration and biomineral formation. IDPs are typically organic components of biominerals. It is believed that they could act as a regulatory coordinator for specific interactions of many proteins, and thus many physiological processes such as formation of dentin and bone, the formation of sea urchin and crusteacean exoskeletons. Here, we review what is currently understood about the molecular basis of biomineral formation. This includes protein interaction with metal ions, post-translational modifications, interactions with other proteins, or other factors that induce the formation of crystal shape and size along with the proper polymorph selection in relation to the role of IDPs.

Molecular aspects of biomineralization of the echinoderm endoskeleton.
Echinoderms possess a rigid endoskeleton composed of calcite and small amounts of occluded organic matrix proteins. The test (i.e., the shell-like structure of adults), spines, pedicellariae, tube feet, and teeth of adults, as well as delicate endoskeletal spicules found in larvae of some classes, are the main skeletal structures. They have been intensively studied for insight into the mechanis...

Fish otoliths, biominerals composed of calcium carbonate with a small amount of organic matrix, are involved in the functioning of the inner ear. Starmaker (Stm) from zebrafish (Danio rerio) was the first protein found to be capable of controlling the formation of otoliths. Recently, a gene was identified encoding the Starmaker-like (Stm-l) protein from medaka (Oryzias latipes), a putative homo...
متن کاملEffect of Phosphorylation on a Human-like Osteopontin Peptide.
The last decade established that the dynamic properties of the phosphoproteome are central to function and its modulation. The temporal dimension of phosphorylation effects remains nonetheless poorly understood, particularly for intrinsically disordered proteins. Osteopontin, selected for this study due to its key role in biomineralization, is expressed in many species and tissues to play a ran...
متن کاملThe Intrinsically Disordered C-RING Biomineralization Protein, AP7, Creates Protein Phases That Introduce Nanopatterning and Nanoporosities into Mineral Crystals
We report an interesting process whereby the formation of nanoparticle assemblies on and nanoporosities within calcite crystals is directed by an intrinsically disordered C-RING mollusk shell nacre protein, AP7. Under mineralization conditions, AP7 forms protein phases that direct the nucleation of ordered calcite nanoparticles via a repetitive protein phase deposition process onto calcite crys...

With the increasing need to integrate different areas of science in the study of intrinsically disordered proteins we arranged a meeting entitled “Intrinsically Disordered Proteins: Connecting Computation, Physics and Biology” in Zürich in September 2013. The aim of the meeting was to bring together scientists from a range of disciplines to provide a snapshot of the field, as well as to promote...
