Pressing mitochondrial genetics forward.

چکیده
Mitochondria are crucial for many cellular functions. In this issue of Cell Reports, studies from Lanning et al. and Wolf and Mootha describe RNAi approaches to screening the mitochondrial proteome. Unexpectedly, they uncover key roles for two poorly characterized mitochondrial proteins: AK4 and FASTKD4. These studies provide examples of the power of forward genetic screens, even when screening a subset of genes, in deciphering functions of previously mysterious mitochondrial proteins.

Positive and negative selection on the mitochondrial genome.
Several recent studies have confirmed that mitochondrial DNA variation and evolution are not consistent with the neutral theory of molecular evolution and might be inappropriate for estimating effective population sizes. Evidence for the action of both positive and negative selection on mitochondrial genes has been put forward, and the complex genetics of mitochondrial DNA adds to the challenge...
متن کاملA forward genetic screen identifies mutants deficient for mitochondrial complex I assembly in Chlamydomonas reinhardtii.
Mitochondrial complex I is the largest multimeric enzyme of the respiratory chain. The lack of a model system with facile genetics has limited the molecular dissection of complex I assembly. Using Chlamydomonas reinhardtii as an experimental system to screen for complex I defects, we isolated, via forward genetics, amc1-7 nuclear mutants (for assembly of mitochondrial complex I) displaying redu...
متن کاملCisplatin cytotoxicity is dependent on mitochondrial respiration in Saccharomyces cerevisiae
Objective(s): To understand the role of mitochondrial respiration in cisplatin sensitivity, we have employed wild-type and mitochondrial DNA depleted Rho0 yeast cells. Materials and Methods: Wild type and Rho0 yeast cultured in fermentable and non-fermentable sugar containing media, were studied for their sensitivity against cisplatin by monitoring growth curves, oxygen consumption, pH changes ...
متن کاملContrasting patterns of nonneutral evolution in proteins encoded in nuclear and mitochondrial genomes.
We report that patterns of nonneutral DNA sequence evolution among published nuclear and mitochondrially encoded protein-coding loci differ significantly in animals. Whereas an apparent excess of amino acid polymorphism is seen in most (25/31) mitochondrial genes, this pattern is seen in fewer than half (15/36) of the nuclear data sets. This differentiation is even greater among data sets with ...
متن کاملConcept for estimating mitochondrial DNA haplogroups using a maximum likelihood approach (EMMA)☆
The assignment of haplogroups to mitochondrial DNA haplotypes contributes substantial value for quality control, not only in forensic genetics but also in population and medical genetics. The availability of Phylotree, a widely accepted phylogenetic tree of human mitochondrial DNA lineages, led to the development of several (semi-)automated software solutions for haplogrouping. However, current...
