چکیده

[2-H] Myoinositol accumidates in rabbit ciliary body-iris preparations in vitro against a concentration gradient. The transport system is energy and temperature dependent (Ql0 = 2.3) and demonstrates saturation kinetics (Km = 0.9 mM.). It is inhibited by ouabain, iodoacetate, phlorhizin, and shows requirements for Na, K+, and Ca. The transport system appears highly specific; various amino acids, ascorbic acid, glucoascorbic acid, and sugar alcohols were without effect on the accumulation of inositol. The stereoisomer scylloinositol was found to inhibit the transport of myoinositol. Dand x.-chiroinositols had no effect. The phosphate esters of inositol competitively inhibited transport of myoinositol. The steady state concentrations of inositol in the lens, ciliary body-iris, aqueous humors, and plasma were determined. The concentration in posterior aqueous was higher than in the anterior aqueous and still higher than in plasma. Evidence is presented to show that the transport of inositol across the blood-aqueous barrier in vivo is a carrier-mediated process and it is suggested that the high concentration of inositol in the aqueous humors is the result of active transport from plasma into posterior aqueous.

Intraocular transport of myoinositol. II. Accumulation in the rabbit lens in vitro.

The characteristics of myoinositol transport in the lens were studied by culturing them in a medium containing the tritium-labeled compound. The accumulation in the lens takes place against a concentration gradient and is inhibited by ouabain, iodoacetate, phlorhizin, and at reduced temperature (Q,o = 3). The energy for transport is derived from glycolysis. The transport system follows Michaeli...

Basolateral inositol transport by intestines of carnivorous and herbivorous teleosts.

Myoinositol transport by isolated basolateral membrane vesicles was characterized from intestines of the herbivorous tilapia (Oreochromis mossambicus) and the carnivorous eel (Anguilla anguilla). [3H]myoinositol transport occurred by nonelectrogenic, facilitated diffusion independent of cation gradients and was inhibited by phloretin (Ki = 0.6 and 0.9 mM for tilapia and eel, respectively) but n...

Hypertonic stress increases NaK ATPase, taurine, and myoinositol in human lens and retinal pigment epithelial cultures.

PURPOSE Recent evidence suggests that taurine and myoinositol may serve as organic osmolytes in a number of cells, including lens and retinal pigment epithelia, but the mechanism for their increased accumulation in response to hypertonic stress is not known. To assess whether NaK ATPase contributed to the elevated levels of taurine and myoinositol in cells exposed to hypertonic media, we measur...

Effect of myoinositol on skin cholesterol levels in rats maintained on a low protein, myoinositol deficient diet.

For sometime past we have been interested in the effect of myoinositol on cholesterol me tabolism in rats (1-3). All these studies were made under experimental conditions designed at this laboratory, which are suitable to investi gate myoinositol deficiency and supplementation. Under such conditions, it has been shown that myoinositol promotes the recovery of delayed transport of hepatogenic ch...

Brain uptake of myoinositol after exogenous administration.

An acute increase in plasma tonicity results in an adaptive increase in brain organic osmolyte content, but this process requires several days to occur. Slow reaccumulation of brain organic osmolytes may contribute to osmotic demyelination. It was investigated whether administration of intravenous myoinositol in rats could speed entry of the osmolyte into the brain. Two groups of animals were s...
