The binding of 2-acetylaminofluorene to rat liver ribonucleic acid in vivo.

چکیده
including the potent hepatic carcinogen, 2-acetylamino fluorene (2-AAF), become bound to tissue protein in susceptible animals (6, 20, 35). Recently, it has been reported (18) and confirmed (10, 19, 38) that radioactive 2-AAF also binds to liver ribonucleic acid (RNA) when administered to rats. Because of its possible importance to our understanding of the pathogenesis of liver cancer, some aspects of the nature of the interaction between 2AAF and liver RXA and of its possible significance in the over-all process of carcinogenesis have been studied. The results of this investigation are reported here.

On the significance and mechanism of the binding of 2-acetylaminofluorene and N-hydroxy-2-acetylaminofluorene to rat-liver ribonucleic acid in vivo.
Previous reports that radioactivity was bound to liver RNA after administration of 2-acetylaminofluorene-9-14C or N-hydroxy-2-acetylaminofluorene-9-14C in species susceptible to the carcinogenic action of 2-acetylaminofluorene have been con firmed. Injection of l-hydroxy-2-acetylaminofluorene-l-14C, a noncarcinogenic metabolite of 2-acetylaminofluorene, did not result in any binding of radioact...
متن کاملCovalent binding of 2-acetylaminofluorene, 2-aminofluorene, and N-hydroxy-2-acetylaminofluorene to rat liver nuclear DNA and protein in vivo and in vitro.
Binding of the hepatocarcinogen 2-acetylaminofluorene (AAF) and two metabolites, 2-aminofluorene (AF) and N-hydroxy-2-acetylaminofluorene (N-OH-AAF), to the DNA and protein of rat hepatic nuclei was examined in vitro and in a cell-free system. Three and one-half hr following a single injection of each compound in equimolar amounts. DNA contained approximately 50% more of the compounds per mg th...
متن کاملEffect of glutathione depletion on the hepatotoxicity and covalent binding rat liver macromolecules of N-hydroxy-2-acetylaminofluorene.
Glutathione plays an important role in the protection of the liver against several hepatotoxins. The hepatocarcinogen N-hydroxy-2-acetylaminofluorene is converted in the rat in vivo to reactive metabolites that bind covalently to cellular macromolecules. These metabolites may also react with glutathione, resulting in the formation of glutathione conjugates and in the detoxification of reactive ...
متن کاملPersistent binding of 2-acetylaminofluorene to rat liver DNA in vivo and consideration of the mechanism of binding of N-hydroxy-2-acetylaminofluorene to rat liver nucleic acids.
Previous reports that radioactivity was bound to rat liver DNA in vivo after administration of 2-acetylaminofluorene (AAF)-9-14C were confirmed. In addition, administration of several other AAF derivatives, namely N-hydroxy-AAF-9 -14C, N-acetoxy-AAF-914C, and the glucuronide of N-hydroxyAAF-9)4C, led to the binding of higher levels of radioactivity to rat liver ribosomal RNA and DNA. Maximum le...
متن کاملBinding of aromatic amines to the rat hepatic Ah receptor in vitro and in vivo and to the 8S and 4S estrogen receptor of rat uterus and rat liver.
Studies on structurally related aromatic amines with different carcinogenic properties have shown that 2-acetylaminofluorene (2-AAF) and 2-acetylaminophenanthrene (AAP) inhibit the binding of 2,3,7,8-tetrachlorodibenzo-p-dioxin to the Ah receptor in vitro. The apparent inhibitor constants (Ki) are 2.3 microM for 2-AAF and 2.7 microM for AAP. In contrast, 4-acetylaminofluorene, an isomer of 2-AA...
