Hepta-, hexa-, and pentacarboxylic porphyrins of porphyria cutanea tarda. II. Preparation of the porphyrins by stepwise decarboxylation ofuroporphyrins.

چکیده
Hepta-, hexa-, and pentacarboxylic porphyrins have been detected in porphyric as well as normal urine samples. From the former they have been isolated and converted into pure methyl esters (1). For a better understanding of the chemistry of these porphyrins, pure uroporphyrin I and uroporphyrin III were separately subjected to decarboxylation under controlled conditions. The decarboxylation products are described and their relations with those isolated from the porphyric urine are discussed in this report.

Porphyrin patterns in different types of porphyria.
Porphyrins from 38 cases of different types of porphyria were separated chromatographically and analyzed. Patients with different types of porphyria eliminated porphyrins with different patterns. Simple paper chromatography of the methyl ester of the total porphyrin of a urine sample revealed a chromatographic pattern of the porphyrins characteristic of the type of porphyria. The relative abund...
متن کاملA high-speed liquid-chromatographic method for measuring urinary porphyrins.
We describe a rapid quantitative and qualitative "high-performance" liquid-chromatographic (HPLC) method for measuring porphyrins in urine. Direct injection of acidified, filtered urine onto a 3-micron (particle size) 3-cm-long reversed-phase column fully resolves uroporphyrin, hepta-, hexa-, and pentacarboxylic acid porphyrins, and coproporphyrin. Instrument response is linearly related to con...
متن کاملPorphyrins and Chlorins from a Congenital Porphyria Patient.
Analysis of porphyrins from the urine, blood, and feces of a congenital porphyria patient showed coproand uroporphyrins from the blood contained more than 95% of the Type I isomer, and those from stool and urine, about 90%. The intermediate hepta-, hexaand pentacarboxylic porphyrins from the urine contained 70% or less of the Type I isomer. The results are discussed and compared with those in p...
متن کاملThe effects in vivo of mutationally modified uroporphyrinogen decarboxylase in different hem12 mutants of baker's yeast (Saccharomyces cerevisiae).
Nine new hem12 haploid mutants of baker's yeast (Saccharomyces cerevisiae), totally or partially deficient in uroporphyrinogen decarboxylase activity, were subjected to both genetic and biochemical analysis. The mutations sites studied are situated far apart within the HEM12 gene located on chromosome IV. Uroporphyrinogen decarboxylase activity in the cell-free extracts of the mutants was decre...
متن کاملSeparation and characterization of pentacarboxylic porphyrinogen isomers by high-performance liquid chromatography with electrochemical detection.
A reversed-phase h.p.l.c. system is described for the separation of all five naturally occurring pentacarboxylic porphyrinogen isomers. The compounds are detected electrochemically with high sensitivity. The peaks are positively identified by h.p.l.c. analysis of the pentacarboxylic porphyrinogens from reduction of pentacarboxylic porphyrins prepared by partial decarboxylation of hexa- and hept...
