چکیده

Recently it was observed that the DNA repair protein human O(6)-alkylguanine-DNA alkyltransferase repairs lesions at the 5' ends of 70-nucleotide single-stranded DNA roughly threefold more frequently than lesions at the 3' ends. Here, we introduce a coarse-grained model to show how a local asymmetry in binding kinetics (rather than thermodynamics) together with irreversible alkyl transfer can give rise to this apparent bias in sequence scanning. Exploration of the parameter space provides quantitative relationships that can be used to validate the proposed mechanism by gel-based assays.

DNA Quality Control by a Lesion Sensor Pocket of the Xeroderma Pigmentosum Group D Helicase Subunit of TFIIH

BACKGROUND Nucleotide excision repair is a versatile DNA repair reaction that removes bulky adducts generated by environmental mutagens such as the UV spectrum of sunlight or chemical carcinogens. Current multistep models of this excision repair pathway accommodate its broad substrate repertoire but fail to explain the stringent selectivity toward damaged nucleotides among excess native DNA. To...

Studies on electron beam induced DNA damage and repair kinetics in lymphocytes by alkaline comet assay

Background: Exposure to ionizing radiation is known to induce oxidative stress followed by damage to critical biomolecules like lipids, proteins and DNA through radiolysis of cellular water. Since radiation has been widely used as an important tool in therapy of cancer, the detailed investigation regarding the DNA damage and repair kinetics would help to predict the radiation sensitivity of cel...

Comparative enzymology of ultraviolet-induced DNA repair synthesis and semiconservative DNA replication in permeable diploid human fibroblasts.

In nongrowing mammalian cells, DNA repair synthesis following irradiation with high doses of UV is almost totally inhibited by aphidicolin, an agent specific for DNA polymerase alpha, and presumably is mediated by that polymerase. In this paper, several enzymologic characteristics of DNA repair synthesis induced in permeable confluent diploid human fibroblasts by high doses of UV have been exam...

Radiosensitivity and Repair Kinetics of Gamma-Irradiated Leukocytes from Sporadic Prostate Cancer Patients and Healthy Individuals Assessed by Alkaline Comet Assay

Background: Impaired DNA repair mechanism is one of the main causes of tumor genesis. Study of intrinsic radiosensitivity of cancer patients in a non-target tissue (e.g. peripheral blood) might show the extent of DNA repair deficiency of cells in affected individuals and might be used a predictor of cancer predisposition. Methods: Initial radiation-induced DNA damage (ratio of Tail DNA/Head DN...

The Role of Long Non Coding RNAs in the Repair of DNA Double Strand Breaks

DNA double strand breaks (DSBs) are abrasions caused in both strands of the DNA duplex following exposure to both exogenous and endogenous conditions. Such abrasions have deleterious effect in cells leading to genome rearrangements and cell death. A number of repair systems including homologous recombination (HR) and non-homologous end-joining (NHEJ) have been evolved to minimize the fatal effe...
