Novel Molecular Assay and Sample Preparation Method for the Detection of Alicyclobacillus in Fruit Juice Concentrates and Bottling Process Materials

چکیده

Figure 1: GeneDisc Plate for TAB Spoilage Alicyclobacilli are thermophilic, acidophilic and spore-forming bacteria (TAB) capable of surviving extreme conditions during the fruit juice manufacturing and bottling processes. These spoilage organisms can generate strong off-flavors and off-odors, which negatively affect product quality in fruit juices, concentrates and ingredients used in beverage production. Though non-pathogenic, they pose a major economic risk to processors. Rapid detection and identification of TAB spoilage at various identified critical control points of the production process is a valuable tool to facilitate release of high quality products.

Detection of Alicyclobacillus spp. in Fruit Juice by Combination of Immunomagnetic Separation and a SYBR Green I Real-Time PCR Assay

An approach based on immunomagnetic separation (IMS) and SYBR Green I real-time PCR (real-time PCR) with species-specific primers and melting curve analysis was proposed as a rapid and effective method for detecting Alicyclobacillus spp. in fruit juices. Specific primers targeting the 16S rDNA sequences of Alicyclobacillus spp. were designed and then confirmed by the amplification of DNA extrac...

A G-quadruplex DNAzyme-based colorimetric method for facile detection of Alicyclobacillus acidoterrestris.

The rapid and sensitive detection of Alicyclobacillus acidoterrestris (AA) has become very important due to the frequent occurrence of fruit juice spoilage by AA. In the present study, using guaiacol, both as the metabolic product of AA related to its concentration and as a green colorimetric substrate of G-quadruplex DNAzyme, a novel G-quadruplex DNAzyme-based colorimetric method for a rapid d...

EVALUATION OF THE DISTRIBUTION AND ACCUMULATION OF SPECIES OF ALICYCLOBACILLUS IN THE FRUIT CONCENTRATE PROCESSING ENVIRONMENT CATHARINA ELIZABETH STEYN Thesis presented in partial fulfilment of the requirements for the degree of MASTER OF SCIENCE IN FOOD SCIENCE

Alicyclobacillus species are thermo-acidophilic bacteria that produce highly resistant endospores able to survive the processing temperatures of fruit concentrate manufacturing, including evaporation and conventional pasteurisation (86 ° 96 °C for ± 2 min). Alicyclobacillus endospores retain their viability in juice concentrates which, under favourable conditions, could germinate and multiply t...

Rapid discrimination of Alicyclobacillus strains in apple juice by Fourier transform infrared spectroscopy.

Alicyclobacillus spp. are thermoacidophilic, spore-forming bacteria. Some of which cause spoilage in pasteurized and heat-treated apple juice products through the production of guaiacol. Fourier transform infrared (FT-IR) spectroscopy was used to discriminate between eight Alicyclobacillus strains (WAC, 81-2, Oly#21, 51-1, KF, 1016, 1101, and A-Gala A4) in apple juice. FT-IR vibrational combina...

Fruit juice and puree characteristics influence enrichment requirements for real-time PCR detection of Alicyclobacillus acidoterrestris

Background: Alicyclobacillus acidoterrestris is a key spoilage causing bacterium commonly found in fruit juices and purees. Commercial real-time PCR based assays to detect this organism are available, but reportedly require 48 hours of enrichment for detection. The underlying hypothesis of this study was that fruit juice and puree characteristics influence the enrichment requirements of this or...
