Adult Stem Cell Plasticity Defined

چکیده
Introduction Investigators have postulated since the turn of the century that some cells in the body appear to participate in the formation of new cells to replace injured, aged, or infected cells in tissues and organs. The concept that certain specialized cells within an organ (stem cells) could give rise to mature functioning cells in that same organ has been demonstrated most clearly in the hematopoietic system. Evidence for stem cell function in the hematopoietic system has come from controlled laboratory experiments and as a consequence of human exposure to massive irradiation during armed conflict. World War II was brought to an end dramatically in August 1945 with the dropping of atomic bombs on the cities of Hiroshima and Nagasaki, Japan. In the wake of the devastation and human suffering of these events, several hundred thousand Japanese civilians and soldiers were exposed to lethal and sublethal doses of irradiation. Examination of the affected people included, but was not limited to, the ablative effects of irradiation on the body’s hematopoietic system. Observation and treatment of these patients led to several scientific reports confirming the postulated existence of stem cells that are responsible for the continuous production of all circulating blood cells. Subsequent studies performed on laboratory rodents detailed the detrimental effects of irradiation on the hematopoietic system as well as the ability of bone marrow transplantation to correct such anomalies. Hematopoietic repopulation was established by transplanting clonogenic marrow cells, which could be tracked to the spleen of the irradiated recipient following infusion. Thus, a single stem cell (clone) from the bone marrow could give rise to thousands of mature blood cells inside the splenic microenvironment within 8 days following transplantation. Over the past 30 years, additional animal and human studies have demonstrated the feasibility of successful transplantation of marrow stem cells into recipients, with reconstitution of the host blood cells with donor type cells and amelioration of congenital or acquired blood disorders. Today we recognize that numerous adult stem cells operate in various capacities in postnatal humans, regulating

High neuronal/astroglial differentiation plasticity of adult rat hippocampal neural stem/progenitor cells in response to the effects of embryonic and adult cerebrospinal fluids
Hippocampal neural stem/progenitor cells (hipp-NS/PCs) of the adult mammalian brain are important sources of neuronal and gial cell production. In this study, the main goal is to investigate the plasticity of these cells in neuronal/astroglial differentiations. To this end, the differentiation of the hipp-NS/PCs isolated from 3-month-old Wistar rats was investigated in response to the embryonic...
متن کاملGrowth Kinetics, Characterization, and Plasticity of Human Menstrual Blood Stem Cells
One of the readily available sources of mesenchymal stem cells (MSCs) is menstrual blood-derived stem cells (Men-SCs), which exhibit characteristics similar to other types of MSCs. This study was performed to determine the growth kinetics, plasticity, and characterization of Men-SCs in women. During spring 2014 in the southern Iranian city of Shiraz, menstrual blood (5 mL) was obtained from 10 ...
متن کاملPhysiological Plasticity of Neural-Crest-Derived Stem Cells in the Adult Mammalian Carotid Body
Adult stem cell plasticity, or the ability of somatic stem cells to cross boundaries and differentiate into unrelated cell types, has been a matter of debate in the last decade. Neural-crest-derived stem cells (NCSCs) display a remarkable plasticity during development. Whether adult populations of NCSCs retain this plasticity is largely unknown. Herein, we describe that neural-crest-derived adu...
متن کاملAdult stem cell plasticity: fact or artifact?
There has been unprecedented recent interest in stem cells, mainly because of the hope they offer for cell therapy. Adult stem cells are an attractive source of cells for therapy, especially in view of the recent claims that they are remarkably plastic in their developmental potential when exposed to new environments. Some of these claims have been either difficult to reproduce or shown to be m...

Objective(s): In the past few decades, variety of foetal, embryonic and adult stem and progenitor cells have been tried with conflicting outcome for cell therapy of central nervous system injury and diseases. Cellular characteristics and functional plasticity of Globose basal stem cells (GBCs) residing in the olfactory epithelium of rat olfactory mucosa have not been studied in the past by the ...
متن کاملFurther proof of the plasticity of adult stem cells and their role in tissue repair
In this issue, De Bari et al. (2003) present elegant data to counter the recent claims that adult stem cells have a limited plasticity. Further, they provide evidence that adult stem cells can seek out damaged tissues and repair them.
