چکیده

microtubule-kinesin system and the actin-myosin system Both systems operate on the same principle; myosin or kinesin motor heads drive specific polymer filaments ma up of protein monomers (actin or tubulin) while utilizing ATP as a chemical energy source (Figure 1 [1]). Actin monomers have an average length of 5.5 nm and join together to form twisted-rope chains 10 nm in diamet with lengths of 50-200 nm. The filaments associated wit kinesin, hollow microtubules, are significantly larger at nm in diameter and up to 100 μm long, usually composed 13 parallel strands of 8 nm long tubulin monomers. Whil they both perform the same basic function, there are many variations among kinesin and myosin molecules, includi head size, hinge length, and mode of motion.

Bidirectional cooperative motion of myosin-II motors on actin tracks with randomly alternating polarities

The cooperative action of many molecular motors is essential for dynamic processes such as cell motility and mitosis. This action can be studied by using motility assays in which the motion of cytoskeletal filaments over a surface coated with motor proteins is tracked. In previous studies of actin-myosin II systems, fast directional motion was observed, reflecting the tendency of myosin II moto...

Smooth and skeletal muscle actin are mechanically indistinguishable in the in vitro motility assay.

Smooth muscle produces as much stress as skeletal muscle with less myosin. To determine if the actin isoforms specific to smooth muscle contribute to the enhanced force generation, the motility of actin filaments from smooth and skeletal muscle were compared in an in vitro assay in which single fluorescently labeled actin filaments slide over a myosin-coated coverslip. No difference was observe...

Myosin-V Opposes Microtubule-Based Cargo Transport and Drives Directional Motility on Cortical Actin

Intracellular transport is driven by motor proteins that either use microtubules or actin filaments as their tracks, but the interplay between these transport pathways is poorly understood. Whereas many microtubule-based motors are known to drive long-range transport, several actin-based motors have been proposed to function predominantly in cargo tethering. How these opposing activities are in...

The effects of smooth muscle caldesmon on actin filament motility.

The movement of reconstituted thin filaments over an immobilized surface of thiophosphorylated smooth muscle myosin was examined using an in vitro motility assay. Reconstituted thin filaments contained actin, tropomyosin, and either purified chicken gizzard caldesmon or the purified COOH-terminal actin-binding fragment of caldesmon. Control actin-tropomyosin filaments moved at a velocity of 2.3...

The tail of myosin reduces actin filament velocity in the in vitro motility assay.

It has been observed that heavy meromyosin (HMM) propels actin filaments to higher velocities than native myosin in the in vitro motility assay, yet the reason for this difference has remained unexplained. Since the major difference between these two proteins is the presence of the tail in native myosin, we tested the hypothesis that unknown interactions between actin and the tail (LMM) slow mo...
