چکیده

There has been considerable debate on the ability of Lactobacillus reuteri to produce the active form of vitamin B12. In this study, we demonstrated the ability of two wild type strains of Lactobacillus reuteri to produce α-(5,6-dimethylbenzimidazolyl)-cobamidcyanide or cyanocobalamin which are the active form of vitamin B12. This was possible by supplying the required compounds of vitamin B12, δ-aminolevulinic acid (ALA) and 5,6-dimethylbenzimidazole (DMB) under the appropriate conditions for bacteria to induce and complete the formation of vitamin B12 with DMB as the lower (Coα) ligand. It was remarkable to find a microorganism possessing probiotic properties along with the capability to produce an active vitamin B12. High performance liquid chromatography (HPLC) coupled with mass spectrometry were used to identify vitamin B12 forms and confirm our results.

Wheat and Sugar Beet Coproducts for the Bioproduction of 3-Hydroxypropionic Acid by Lactobacillus reuteri DSM17938

An experimental design based on Response Surface Methodology (RSM) was used for the formulation of a growth medium based on sugar beet and wheat processing coproducts adapted to the cultivation of Lactobacillus reuteri (L. reuteri) DSM17938. The strain was cultivated on 30 different media varying by the proportions of sugar beet and wheat processing coproducts, and the concentration of yeast ex...

Functional identification in Lactobacillus reuteri of a PocR-like transcription factor regulating glycerol utilization and vitamin B12 synthesis

BACKGROUND Lactobacillus reuteri harbors the genes responsible for glycerol utilization and vitamin B12 synthesis within a genetic island phylogenetically related to gamma-Proteobacteria. Within this island, resides a gene (lreu_1750) that based on its genomic context has been suggested to encode the regulatory protein PocR and presumably control the expression of the neighboring loci. However,...

Heterologous expression of a recombinant lactobacillal β-galactosidase in Lactobacillus plantarum: effect of different parameters on the sakacin P-based expression system

BACKGROUND Two overlapping genes lacL and lacM (lacLM) encoding for heterodimeric β-galactosidase from Lactobacillus reuteri were previously cloned and over-expressed in the food-grade host strain Lactobacillus plantarum WCFS1, using the inducible lactobacillal pSIP expression system. In this study, we analyzed different factors that affect the production of recombinant L. reuteri β-galactosida...

BluB/CobT2 fusion enzyme activity reveals mechanisms responsible for production of active form of vitamin B12 by Propionibacterium freudenreichii

BACKGROUND Propionibacterium freudenreichii is a food grade bacterium that has gained attention as a producer of appreciable amounts of cobalamin, a cobamide with activity of vitamin B12. Production of active form of vitamin is a prerequisite for attempts to naturally fortify foods with B12 by microbial fermentation. Active vitamin B12 is distinguished from the pseudovitamin by the presence of ...
